摘要
以不同蛋白酶酶解制备家蚕蛹蛋白酶解产物(SPPHs),采用MTT比色法检测其对人胃腺癌细胞MGC-803的增殖抑制活性。不同蛋白酶酶解制备SPPHs对MGC-803的增殖抑制作用存在显著差异(P〈0.05),其中家蚕蛹蛋白以Alcalase 2.4L酶解的产物(AH)对MGC-803的增殖抑制活性最高,当AH的质量浓度为0.25 mg/m L时,水解度(DH)=25%的AH的抑制活性最高,对MGC-803细胞的增殖抑制率可达到92.74%。Alcalase2.4L酶解家蚕蛹蛋白产物的分子质量也显著影响其对MGC-803细胞的增殖抑制活性,其中分子质量〈5 k D组分的抑制率高达91.13%,将该组分经葡聚糖(G-25)凝胶层析后分离出4个组分(Ⅰ、Ⅱ、Ⅲ、Ⅳ),组分Ⅱ对MGC-803细胞的增殖抑制率最高(27.5%),但与未分离样品相比,抑制效率显著降低,而其他3个分离组分则无抑制作用。研究结果提示,蚕蛹蛋白酶解产物对肿瘤细胞的体外增殖有抑制作用,并且这种抑制作用可能是由多种组分协同完成。
Proliferation inhibitory effect of silkworm pupa protein hydrolysates( SPPHs) on gastric cancer cell MGC-803 was investigated in by MTT colorimetric assay. Proliferation inhibitory effect of SPPHs prepared by different enzyme on MGC-803 cell was significantly different. The proliferation inhibitory effect of Alcalase2. 4 L hydrolysates( AH) was the best( 92. 74%) when its degree of hydrolysis was 25%. Molecular weight of AH can also influence the inhibitory effect of proliferation in MGC-803 cell. The component of molecular weights less than 5 k D had an inhibitory effect of 91. 13%on tumor cell proliferation. Four factions( Ⅰ,Ⅱ,Ⅲ andⅣ) were separated from this component by Sephadex gel chromatography( G-25). The proliferation inhibitory effect of faction Ⅱ on MGC-803 tumor cells was the highest( 27. 59%),but the proliferation inhibitory effect was significantly decreased as compared with the unseparated sample,and the other three fractions had no proliferation inhibitory effect. Our results suggest that the enzymatic hydrolysates of silkworm pupa proteins can inhibit the proliferation of tumor cells in vitro,and the inhibition maybe accomplished by synergistic action of multiple components.
