摘要
目的 探究全身过敏反应对ApoE-/-C57BL/6小鼠动脉粥样硬化(atherosclerosis,AS)斑块以及肥大细胞数量的影响.方法 15只ApoE-/-C57BL/6小鼠髙脂饮食喂养后随机分成三组:对照组、10μg卵清蛋白(ovalbumin,OVA)免疫组和50μg OVA免疫组.激发过敏反应2 h后处死小鼠,取血取材.异种被动皮肤过敏(passive cutaneous anaphylaxis,PCA)实验检测免疫球蛋白(immunoglobulin,Ig)相对含量,小鼠主动脉HE染色观察AS斑块大小;甲苯胺蓝染色法观察肥大细胞数量.结果 OVA激发使小鼠发生过敏反应,体温明显下降,且50μg剂量组体温下降程度明显大于10μg剂量组;OVA激发过敏反应组的PCA试验为阳性结果,且50μg剂量组蓝斑直径大于10ug剂量组,差异有统计学意义[(8.10±0.60)比(4.20±0.20),t=6.17,P〈0.05];10μg剂量组的小鼠斑块大小基本无变化,50μg剂量组的斑块大于对照组,差异有统计学意义[(0.53±0.04)比(0.30±0.02),t=4.95,P〈0.05];对照组和10μg剂量组肥大细胞的数量差别不大(P〉0.05);50μg剂量组相对对照组可观察到较多数的肥大细胞,差异有统计学差异[(21.20±2.33)比(3.75±1.44),t=5.96,P〈0.05].结论 不同浓度的OVA作用ApoE-/-C57BL/6小鼠时可使其产生不同程度的过敏反应,出现较重过敏症状的ApoE-/-C57BL/6小鼠动脉粥样硬化血管有较多的肥大细胞聚集.
Objectives To investigate the anaphylactic impact on the development of aortic atheroscle-rosis and the recruitment of mast cells. Methods Fifteen ApoE-/ - C57BL/ 6 mice were fed on western diet and divided them randomly into three groups:the control groups,the group immuned by 10 μg OVA,and the group immuned by 50 μg OVA. The blood and tissues were sampled in 2 h after anaphylaxis was triggered. Then we conduct the experiment of xenogenic passive cutaneous anaphylaxis(PCA)to detect the relative content of IgE. Hematoxylin and eosin staining was performed to observe the plaque size on the aortic tissue,and Toluidine blue staining was performed to observe the mast cell number in the atherosclerotic plaque. ResuLts The decrease of body temperature were observed after triggered of OVA,and the responses were more pronounced in the group of 50 μg OVA. The PCA experiment was positve in the groups of OVA. The diameter of dermal Evan's blue spot in the group of 50 μg OVA was larger than that in the group of 10 μg OVA,the difference was statistically signifi-cant[(8. 10 ± 0. 60)vs (4. 20 ± 0. 20),t = 6. 17,P 〈 0. 05]. The sizes of atherosclerotic plaque had no changed in the group of 10 μg OVA. Anaphylaxis triggered by 50 μg OVA can significantly increased athero-sclerotic plaque compared with the control group,the difference was statistically significant[(0. 53 ± 0. 04)vs (0. 30 ± 0. 02),t = 4. 95,P 〈 0. 05]. The number of mast cells in control group and the group of 10 μg OVA had no significant difference(P 〉 0. 05). And the number of mast cells in the group of 50 μg OVA was larger than that in the control group of,the difference was statisticallyt[(21. 20 ± 2. 33)vs (3. 75 ± 1. 44),t = 5. 955, P 〈 0. 05]. . Conclusion The different degrees of anaphylacitic reaction of ApoE-/ - mice can be triggered by different concentration of OVA,and the mast cells will trend to recruit in anaphylactic vessel in those ApoE-/ -C57BL/ 6 mice with more serious symptoms.
