摘要
目的了解北京市门头沟区腮腺炎病毒(Mumps Virus,MuV)的基因特征。方法采集腮腺炎患者咽拭子标本分离病毒,采用逆转录-聚合酶链反应(RT-PCR)扩增小疏水蛋白(small hydrophobic protein,SH)基因片段,对扩增产物进行序列测定和基因分型。结果门头沟区MuV分离株属于F基因型,与2011年香港流行株最为接近。北京不同年代的MuV流行株分散在亲缘关系树不同的进化分支上,无明显的时间和地理分布趋势。与F基因型参考株比对,与浙江株核苷酸的同源性为97.2%,氨基酸的同源性为93.1%;与中国疫苗株JERYL-LYNN核苷酸差异达到19.0%。结论该分离株为F基因型腮腺炎病毒,与北京近年流行株隶属不同的传播链。SH蛋白氨基酸一些保守位点已发生变异,因此应加强MuV基因型监测。
[Objective] To investigate the genetic characteristics of mumps virus(Mu V) in Mentougou District of Beijing.[Methods] Specimens of throat swabs from mumps patients were collected for the isolation of mumps virus. The small hydrophobic protein(SH) gene of isolated strains was amplified by the reverse transcription-polymerase chain reaction(RT-PCR),on which gene sequence and genotype were conducted. [Results] Isolated strain was F genotype which was closest to the epidemic strain isolated in Kong Hong in 2011. The mumps virus sequences of Beijing in different years showed dispersed distributions in phylogenetic tree,which was no obvious tendency of the distribution for years and geography. Compared with mumps virus F reference strains,the homology of nucleotide and amino acid of Mentougou Mu V with Zhejiang reference strain was within 97.2% and 93.1% respectively. 19.0% of differences in nucleotide was between the isolated strain and the Chinese vaccine strain JERYL-LYNN. [Conclusion] The isolated strains of virus are genotype F mumps virus,lineages different from recent Beijing strains. It is necessary to strengthen the monitoring of the mumps virus genotypes because some conserved sites of SH protein amino acids have mutated.
