摘要
生长素抑制蛋白(auxin-repressed protein, ARP)和休眠相关基因(dormancy-related proteins, DRM)是植物中常见的生长素抑制类基因,与植物休眠进程相关。为了鉴定葡萄ARP/DRM基因参与休眠的功能特点,本研究基于欧洲葡萄基因组数据库挖掘,采用同源克隆法获得了3条葡萄ARP/DRM基因家族成员序列,分别命名为VvARP/DRM1、VvARP/DRM2和VvARP/DRM3。序列分析的结果表明,所克隆的3条序列与其他植物的相应序列具有很高的一致性。实时荧光定量表达的检测结果表明,在单氰胺打破葡萄冬芽休眠的过程中,3个基因都是在处理前期表达较高,然后随着处理时间的延长表达水平不断下降。在整个单氰胺处理的过程中,对照组中的VvARP/DRM1、VvARP/DRM2和VvARP/DRM3的表达水平一直高于处理组。研究结果为深入分析ARP/DRM基因家族在葡萄冬芽休眠解除过程中的功能和作用机理提供了参考依据。
Auxin-repressed protein(ARP) and dormancy-related proteins(DRM) are common auxin suppressor genes in plants, which are related to the dormancy process of plants. To identify the functional characterization of ARP/DRM gene family involved in dormancy, the study obtained three family members of ARP/DRM gene by homologous cloning based on the European grape genome database, named VvARP/DRM1 VvARP/DRM2 and VvARP/DRM3 respectively. Sequence analysis revealed that 3 sequences cloned showed high consistency with those from other plants. The results of real-time RT-PCR demonstrated that the expression level of 3 genes were higher in early phase of dormancy-release broken by hydrogen cyanamide, then decreased with the progressing time. The expression level of VvARP/DRM1, VvARP/DRM2 and VvARP/DRM3 in control group were always higher than that of in treatment group in whole treatment progress. The results would provide a reference for further analysis of the function and mechanism of ARP/DRM gene family in the process of grape winter bud dormancy release.
