液相色谱串联质谱法测定血清中腺苷含量

摘要 文章以血清为主要实验材料,建立了液相色谱串联质谱法测定血清中腺苷含量。样品经乙腈沉淀蛋白提取净化后,采用液相色谱串联质谱法(HPLC-ESI-MS/MS)分析,多反应监测模式下(MRM)采用外标法定量。实验结果表明,本实验前处理方法方便快捷,选择性好,回收率高,能够满足实验室大量、快速分析血清中腺苷含量的检测,也可用于血清中腺苷浓度监测研究,为临床诊断和生物医学研究提供科学依据。 A new method using of HPLC - ESI - MS / MS for the analysis of Adenosine in serum has beendeveloped. The samples were extracted by ACN and then determined by LC - ESI - MS / MS. Separated analyteswere determined using multiple reaction monitoring (MRM). The result indicated that the calibration curveswere linear in the range of 5 ~ 100μg / L for adenosine. The quantitation limit was 5. 0μg / kg. The recoveries ofcholesterol were in the range of 80. 2 ~ 82. 5%. This method shortened the time of pre - treatment process, andthe result was credible. It could satisfy the requirement of mass and fast analysis in the laboratory.
