摘要
目的探讨利拉鲁肽抑制高糖诱导足细胞损伤的作用机制。方法培养小鼠肾脏足细胞MPC5,按照培养条件分为对照组(Mock)、高糖组(HG,10、20、30、40 mmol/L)和30 mmol/L D-葡萄糖+不同浓度利拉鲁肽组(1、10、50、100 nmol/L)。干预36h后收集足细胞,RT-PCR检测Grem1 mRNA的表达,免疫印迹检测Nephrin表达。结果给予30 mmol/L葡萄糖作用36 h后,Grem1 mRNA表达明显升高,同时Nephrin表达水平降低(P<0.01)。在高糖条件下给予不同浓度的利拉鲁肽,结果表明利拉鲁肽部分逆转高糖上述作用,且存在剂量依赖性。结论利拉鲁肽可抑制高糖诱导的足细胞损伤。
Objective To investigate the mechanism of liraglutide on the podocyte injury induced by high glucose.Methods Mouse kidney podocytes MPC5 were cultured, then were divided into control group (Mock) and high glucosegroup (HG, 10, 20, 30, 40 mmol/L) and 30 mmol/L Dglucose+different concentrations liraglutide group (1, 10, 50, 100nmol/L) according to culture conditions. After 36h of intervention, the podocytes were collected, and the expression ofGrem1 mRNA was detected by RT -PCR, nephrin expression was detected by western blot. Results After 30 mmol/Lglucose treatment for 36 h, the expression of Grem1 mRNA increased significantly, and the levels of Nephrin decreased(P〈0.01) . The results of giving different concentrations of liraglutide under high glucose condition showed that liraglutidepartially reversed the above effects of high glucose in a dosedependent manner. Conclusion Liraglutide can inhibitpodocyte injury induced by high glucose.
