摘要
目的:建立高效液相色谱法(HPLC)测定冬凌草甲素脂质体的含量。方法:色谱柱:Agilent Zorbax C_(18)柱(250 mm×4.6 mm,5μm),流动相:甲醇-水(55∶45),检测波长:241 nm,柱温:30℃,流速:1 mL/min;进样量:20μL。结果:冬凌草甲素在10~100μg/mL之间线性关系良好,r=0.999 5。日内、日间RSD为0.8%和1.2%,重复性RSD为1.92%,方法回收率平均为99.9%,RSD为0.77%,样品的平均包封率为95.0%。结论:该法简便,准确度高,可用于冬凌草甲素脂质体的含量测定。
Objective:A high performance liquid chromatography(HPLC)method was established for the determination of oridonin in ozagrel hydrochloride liposome. Methods:A C(18) column(250 mm ×4.6 mm,5 μm)was used with the mobile phase of methanol-H2O(55 ∶45)at the detection wavelength of 241 nm. The flow rate was at 1.0 mL/min. The column temperature was 30 ℃. 20 μL sample was injected. Result:The linear range of oridonin was 10~100 μg/mL(r=0.999 5).The inter-and intra-day RSD was 0.8%,1.2%. The method recoveries were 99.9%. The average entrapment efficiency of tartrate indoquinoline liposome was 99.9%(RSD=0.77%). Conclusion:This method is simple,accurate and can be used to control the quality of oridonin Liposome.
