星点设计-效应面法优化丝素蛋白微球的制备工艺 被引量:2

Optimization of Preparation Process of Silk Fibroin Microsphere by Central Composite Design-Response Surface Methodology

摘要 目的:以丝素蛋白为原料制备丝素蛋白微球,并以均匀度(Y)为指标筛选出丝素蛋白微球的最佳制备工艺。方法:以丝素蛋白溶液为水相,以液体石蜡为油相,以戊二醛为交联剂,以Span80为乳化剂,应用乳化-化学交联法制备丝素蛋白微球;用星点设计对丝素蛋白微球的制备工艺进行优化。结果:制得的丝素蛋白微球圆整,其粒径在常规粒径范围1~250μm内;用星点设计优化后的方案为:液体石蜡与丝素蛋白溶液的比例为10∶1,液体石蜡与乳化剂的比例为10%,液体石蜡与戊二醛的比例为33.34∶1。结论:以丝素蛋白为原料,制得了丝素蛋白微球,为后期将丝素蛋白微球作为药物载体的研究奠定基础。 Objective: To optimize the preparation process of silk fibroin microspheres with silk fibroin while taking evenness degree as the index. Method: Took silk fibroin as aqueous solution,liquid paraffin as oil phase,glutaraldehyde as cross linking agent,and Span80 as emulsifier to prepare silk fibroin protein microspheres by emulsion chemical crosslinking,and to optimize its prepprocess by using central composite design.Result: The prepared silk fibroin microspheres were round,and the particle size was in the conventional size range of 1 - 250μm. The optimized scheme with the central composite design was as following: the ratio of liquid paraffin and silk fibroin solution was 10∶ 1,the ratio of liquid paraffin and emulsifier was 10%,and the ratio of the proportion of liquid paraffin and glutaraldehyde was 33. 34 ∶ 1. Conclusion: Preparing silk fibroin microspheres with silk fibroin as raw material lay the foundation for take silk fibroin microsphere as drug carrier in later period.
