摘要
以纳米粒子为磁核,天然多聚物壳聚糖为高分子包裹材料,制备出用于提纯凝血酶的新型亲和磁性微球,并对其凝血酶吸附量、重复使用性等进行了研究。结果显示,壳聚糖亲和磁性微球其透光率在4min时达到93%,并在10min内透光率从51%提高至96%;而在无外加磁场时,其透光率仅从52%提高至59%;相比而言,自制的壳聚糖亲和磁性微球在外加磁场下具备较高的磁响应性。壳聚糖亲和磁性微球对凝血酶粗酶液的纯化参数为:纯化倍数13,纯化时间为0.83h,吸附容量为4956U/g;结果表明,壳聚糖亲和磁性微球纯化凝血酶粗酶液效果较好。
Nano-particles and natural polymer chitosans were separately used as magnetic cores and polylner par- eel materials, a novel affinity magnetic microsphere for thormbin purification was prepared, and its thrombin ad- sorption capacity, reusability were detected. The results showed that the transmittance of chitosan-affinity magnetic microspheres reached 93% at 4 minutes, and the transmittance increased from 51% to 96% within 10 minutes. However, when the magnetic field was not applied, it was just increased from 52% to 59%. In comparison, in magnetic field, the self-made chitosan-affinity magnetic microspheres have a strong magnetic response property. The purification parameters of thrombin crude enzyme solution by chitosan-'affinity magnetic microspheres were as follows: purification factor is 13, purification time is 0.83h, adsorption capacity is 4956U/g; the results showed that the chitosan-affinity magnetic microspheres had a good purification effect on the thrombin crude enzyme solution.
