UVC诱导CD4^+T细胞表面CRT分子表达的动力学特征 被引量:1

摘要 目的探讨UVC处理后CD4^+T淋巴细胞表面钙网蛋白(calreticulin,CRT)的分子动力学特征。方法应用免疫磁珠分选技术分选得到人外周血CD4^+T淋巴细胞,采用流式、细胞免疫荧光等方法检测UVC处理后CD4^+T淋巴细胞表面CRT的分子动力学改变,并分析植物血球凝集素(phytohaemagglutinin,PHA)处理后对CD4^+T淋巴细胞表面CRT的分子动力学的影响。结果1)活化的CD4^+T淋巴细胞经UVC处理后细胞表面出现CRT的暴露,并且随着UVC处理时间的延长,CRT的暴露也随之增多。2)经UVC处理后的CD4^+T淋巴细胞出现细胞凋亡现象,且UVC诱导的CRT暴露早于凋亡过程中的磷脂酰丝氨酸的外翻。3)还发现经PHA刺激活化的CD4^+T细胞相对于未活化的细胞而言细胞表面CRT的暴露量更多。结论 UVC处理使CD4^+T淋巴细胞表面出现CRT分子动力学改变的一系类特征为将来进一步研究经UVC灭活后的CD4^+T淋巴细胞诱导机体产生免疫原性反应(T细胞疫苗)的机制提供了一定的理论基础。 To investigate the molecular dynamics of calreticulin (CRT) on CD4^+ T lymphocytes after UVC treatment, immunomagnetic bead sorting technique was used to sort CD4^+ T lymphocytes from human peripheral blood. The molecular dynamics changes of CRT on CD4^+ T lymphocytes were detected by flow cytometry, immunofluorescence and other methods. The effect of phytohaemagglutinin (PHA) on the molecular dynamics of CRT on CD4^+ T lymphocytes was evaluated. Data showed that CRT was exposured on activated CD4^+ T lymphocytes after UVC treatment, and CRT exposure was increased with UVC-irradiation time prolonging. Apoptosis of CD4^+ T lymphocytes appeared after UVC treatment, and UVC-induced CRT exposure was earlier than that of phosphatidylserine during apoptosis. It was also found that CD4^+ T cells activated by PHA showed more CRT exposure than non-activated cells. In conclusion, the characteristics of UVC-induced CRT molecular dynamics on the surface of CD4^+ T lymphocyte provide theoretical basis for further studies on the mechanism of which UVC-inactivated CD4^+ T cells induce the body to produce an immunogenic response (T cell vaccine)
