摘要
为提高药用观赏植物常春油麻藤繁殖速率,以不同月份采集的不同成熟程度的常春油麻藤荚果种子为外植体建立再生体系,并对瓶苗移植后的生长状况进行跟踪观测。研究结果表明:胚芽萌发诱导最优培养基配比为MS+BA 2.0 mg·L^(-1)+NAA 0.5 mg·L^(-1)+KT 1.0 mg·L^(-1)+蔗糖20 g·L^(-1)+活性炭1.5 g·L^(-1)+琼脂6.5 g·L^(-1);茎段芽诱导和增殖培养最优培养基配比为MS+BA 2.0 mg·L^(-1)+NAA 0.3 mg·L^(-1)+KT 0.5 mg·L^(-1)+TDZ 0.02 mg·L^(-1)+蔗糖20 g·L^(-1)+活性炭1.0 g·L^(-1)+琼脂7.5 g·L^(-1);壮苗培养最优培养基配比为MS+IBA 1.0 mg·L^(-1)+BA 1.0 mg·L^(-1)+KT 0.5mg·L^(-1)+NAA 0.3 mg·L^(-1)+蔗糖25 g·L^(-1)+活性炭1.0 g·L^(-1)+琼脂7.0 g·L^(-1);生根培养最优培养基配比为1/2MS+IBA 0.5 mg·L^(-1)+BA 1.0 mg·L^(-1)+NAA 0.5 mg·L^(-1)+蔗糖25 g·L^(-1)+活性炭1.0 g·L^(-1)+琼脂7.5 g·L^(-1)。瓶苗移栽最优基质体积比为泥炭土∶珍珠岩∶蛭石=5∶1∶2,成活率高达90%以上。
In order to improve the propagation rate of medicinal ornamental plant Mucuna sempervirens Hemsl and to promote the sustainable development,the seeds of Mucuna sempervirens Hemsl pods with different maturities were collected as explants and conducted the tissue culture for them,and the growth state of the bottle seedlings was observed. Results showed that,the optimum medium for the germ induction was MS + BA 2. 0 mg·L^(-1)+ NAA 0. 5 mg·L^(-1)+ KT 1. 0 mg·L^(-1)+ sucrose 20 g·L^(-1)+ AC 1. 5 g·L^(-1)+ agar 6. 5 g·L^(-1); the culture medium suitable for stem bud induction and proliferation culture was MS + BA 2. 0 mg·L^(-1)+NAA 0. 3 mg·L^(-1)+ KT 0. 5 mg·L^(-1)+ TDZ 0. 02 mg·L^(-1)+ sucrose 20 g·L^(-1)+ AC 1. 0 g·L^(-1)+ agar 7. 5 g·L^(-1); the optimum medium was MS + IBA 1. 0 mg·L^(-1)+ BA 1. 0 mg·L^(-1)+ KT 0. 5 mg·L^(-1)+ NAA 0. 3 mg·L^(-1)+ sucrose 25 g·L^(-1)+ AC1. 0 g·L^(-1)+ agar 7. 0 g·L^(-1); the optimum culture medium for rooting culture was 1/2 MS + IBA 0. 5 g·L^(-1)+ BA 1. 0 mg·L^(-1)+NAA 0. 5 mg·L^(-1)+ sucrose 25 g·L^(-1)+ AC 1. 0 g·L^(-1)+ agar 7. 5 g·L^(-1); the optimal volume ratio of transplanting substrate was: peat soil∶ perlite∶ vermiculite = 5∶ 1∶ 2,the survival rate could reach more than 90%.
