澳洲茄边碱对大肠癌RKO细胞增殖和凋亡作用

Effects of Solamargine on proliferation and apoptosis in colorectal carcinoma RKO cells

目的观察澳洲茄边碱对人大肠癌RKO细胞增殖及凋亡作用。方法不同浓度澳洲茄边碱作用RKO细胞。CCK-8(Cell Counting Kit-8,CCK-8)法检测细胞增殖,平板法检测克隆形成,Annexin V-APC/PI标记流式细胞仪检测细胞凋亡,试剂盒检测Caspase-3活性。结果澳洲茄边碱可抑制RKO细胞增殖,呈剂量与时间依赖性。澳洲茄边碱可抑制RKO细胞克隆形成,呈剂量依赖性。澳洲茄边碱可促RKO细胞凋亡。澳洲茄边碱同时可增强RKO细胞Caspase-3活性。结论澳洲茄边碱可以抑制RKO细胞增殖和克隆形成,促RKO细胞凋亡,其机制与活化Caspase-3相关。

Objective To observe the effects of Solamargine on cell proliferation and apoptosis in human colorectal carcinoma RKO cells. Methods RKO cells were treated with different dose of Solamargine. Cell proliferation was detected with CCK - 8 （ Cell Counting Kit - 8 ） assay. Clone formation was observed by plate colony forming assay. Apoptosis was detected by Annexin V - APC/PI staining and identified by flow cytometry. Caspase - 3 activity was measured by commercial kit. Results Solamargine inhibited cell proliferation of RKO ceils in a dose - and time - dependent manner. Solamargine inhibited clone formation in a dose -dependent manner. Solamargine induced apoptosis in RKO ceils. Solamargine also activated Caspase -3 in RKO ceils. Conclusion Solamargine inhibits proliferation and clone formation, induces apoptosis in RKO cells, and may relate to activation of Caspase - 3.
