秦巴山区猪苓多糖的提取及其脂质体生产工艺的优化 被引量:3

摘要 优化秦巴山区猪苓多糖的提取及其脂质体的生产工艺,为其工业化生产提供理论依据。采用响应面分析和Design-Expert 8.0.6软件相结合的方法,确定猪苓多糖提取最佳工艺,并对多糖进行抑菌试验;采用逆向蒸发法制备猪苓多糖脂质体,并用L_9(3~3)正交试验对脂质体制备工艺进行优化;采用硫代巴比妥酸法,研究维生素E和C对脂质体抗氧化的影响。猪苓多糖提取的最佳工艺为:浸提次数3次、液料比143∶15(mL·g^(-1))、浸提时间1.3h;在此条件下,多糖提取率达5.19%。猪苓多糖脂质体制备的最佳工艺为:大豆卵磷脂、胆固醇和聚乙二醇3350的摩尔比7.00∶2.00∶0.40,超声时间5.50 min,猪苓多糖添加量为2.00 mL,猪苓多糖脂质体包封率为98.87%,不同剂量的维生素E和C均可提高脂质体的抗氧化性。 The purpose of this study was to optimize the extraction of Polyporus umbellatus polysaccharide in Qinba Mountains and the production process of its liposomes to provide a theoretical basis for industrialized production. Response surface analysis and Design-Expert 8.0.6 software were used to determine the optimum extraction process of Polyporus umbellatus polysaccharide, which antibacterial activity was further tested. Polyporus umbellatus liposomes were prepared by reverse evaporation method and optimized by L9(33) orthogonal test; the thiobarbituric acid method was used to study the effect of vitamin E and C on liposome anti-oxidation. The optimal extraction conditions were achieved and listed as follows: extraction for 3 times, liquid-to-solid ratio of 143∶15 (mL·g-1), extraction time of 1.3 h. Under these conditions, polysaccharide extraction rate was 5.19%. The process for the preparation of Polyporus umbellatus polysaccharide liposome was optimized as: soy lecithin, cholesterol and PEG 3350 at the molar ratio of 7.00∶2.00∶0.40; ultrasonic time, 5.50 min; the amount of polyporus polysaccharide added, 2.00 mL, and the obtained Polyporus umbellatus liposome encapsulation efficiency was 98.87%; different doses of vitamin E and C could improve the liposome anti-oxidation.
