南瓜糖蛋白的提取分离及降糖作用的研究 被引量:3

摘要 本试验采用水提醇沉法和硫酸铵分级沉淀法提取分离南瓜糖蛋白,研究南瓜糖蛋白对糖尿病小鼠的降血糖作用。首先对提取产物进行定性检测;然后用中性蛋白酶酶解提取物;最后建立四氧嘧啶型糖尿病小鼠模型,选取阴性对照组、阳性对照组、低剂量组、高剂量组四组中造模成功的小鼠,分别连续7 d灌胃一定体积生理盐水、格列苯脲(200 mg/kg)、南瓜糖蛋白(200 mg/kg)及南瓜糖蛋白(400 mg/kg),同时监测每天的体重和第2、4、7 d的血糖值。试验结果表明:高剂量组的降糖率为33.75%,低于格列本脲组的39.4%,高于低剂量组的22.4%。因此南瓜糖蛋白对四氧嘧啶型糖尿病小白鼠的体重和血糖有显著影响,高剂量组比低剂量组降糖率高,但低于格列本脲。 This experiment adopts water extraction and alcohol precipitation method and ammonium sulfate fractional precipitation method to extract and separate pumpkin glycoprotein,to Study the extraction and separation of pumpkin glycoproteins and Its Glucatonic effect. Firstly the qualitative detection was carried out on the extracted product.Then pumpkin glycoproteins was hydrolyzed by neutral protease.Finally the alloxan diabetic mice model was established. We selecting the negative control group, positive control group, lowdose group, high dose group of building successful in four groups of mice, lavage for 7 days, respectively, a certain volume of physiological saline, Glenn benzene urea (200 mg/kg), pumpkins glycoprotein (200 mg/kg) and the glycoprotein (400 mg/kg). At the same time every day test weight and blood sugar levels. The test results show that the positive control group made after the success of the mould of a value of 39.4% drop in blood sugar; Low concentration after the success of the treatment group compared with the building of the blood sugar value is 22.4%; High concentration after suc Cess of the treatment the group compared with the building of the blood sugar value of 33.75%. So Pumpkin glycoprotein has a significant effect on the weight and blood sugar of alloxan-type diabetic mice. The blood sugar lowering rate in the high dose group was higher than that in the low dose group,but slightly lower than Glenn benzene urea.
