四物汤对小鼠骨髓基质细胞凋亡及Bim基因表达的影响

Effect of Siwu Decoction on Apoptosis of Bone Marrow Stromal Cells of Mice and Expression of Bim

目的：观察四物汤对小鼠骨髓基质细胞OP9细胞凋亡的影响,检测四物汤作用后促凋亡因子Bim的基因表达,探讨四物汤对OP9细胞凋亡的作用及机制。方法：流式细胞术检测四物汤对OP9细胞凋亡的影响,实时荧光定量PCR、western blot检测Bim基因表达。结果：四物汤显著抑制OP9细胞凋亡,显著下调Bim的基因表达（P〈0.05,P〈0.01）。结论：四物汤具有抑制OP9细胞凋亡的作用,其机制可能与下调Bim的基因表达有关。

Objective：To observe the effect of Siwu decoction on the apoptosis of OP9 cells,bone marrow stromal cells of mice,to detect the expression of proapoptotic factors of Bim after Siwu decoction intervention,and to explore the effect and mechanism of Siwu decoction on the apoptosis of OP9 cells. Methods：The effect of Siwu decoction on apoptosis of OP9 cells was detected by flow cytometry,and the expression of Bim was detected by qRT-polymerase chain reaction（qRT-PCR） and western blot. Results：Siwu decoction inhibited the apoptosis of OP9 cells significantly and down-regulated the expression of Bim significantly.Conclusion：Siwu decoction can inhibit the apoptosis of OP9 cells,and its mechanism may be related to the down-regulation of Bim expression.
