摘要
目的探讨姜黄素(curcumin)对睡眠剥夺小鼠血清心肌酶、氧化应激酶和炎症因子表达的影响。方法将50只C57雄性小鼠随机分为正常对照组、睡眠剥夺模型组和姜黄素低、中、高剂量组。采用改良多平台水环境法(modifid multiple platrorm water environmental method,MMPWM)进行睡眠剥夺连续72 h,姜黄素低、中、高剂量组按50、100、150 mg/kg进行灌胃,正常对照组和睡眠剥夺模型组小鼠在相同时间点给予等体积生理盐水灌胃,连续8周,每天1次。采用BS-220全自动生化分析仪检测血清心肌酶中心肌酶乳酸脱氢酶(lactate dehydrogenase,LDH)、肌酸激酶(creatine kinase,CK)、α-羟丁酸脱氢酶(alpha-hydroxybutyrate dehydrogenase,α-HBDH)、谷草转氨酶(aspartate aminotransferase,AST)含量;黄嘌呤氧化酶法检测心肌组织中超氧化物歧化酶(superoxide dismutase,SOD)活性,比色法检测谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活性,硫代巴比妥酸(thiosulfuric acid barbitone,TBA)法检丙二醛(malonaldehyde,MDA)含量;ELISA法检测心肌组织中白介素-1β(interleukin-1β,IL-1β)、白介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumor necrosis factorα,TNF-α)含量。结果与睡眠剥夺模型组比较,姜黄素各剂量组小鼠血清心肌酶LDH、CK、α-HBDH、AST含量均下降(P<0.000 1);小鼠心肌组织中SOD、GSH-Px活性逐渐增强(P<0.05),MDA含量逐渐下降(P<0.05),IL-1β、IL-6、TNF-α含量均逐渐下降(P<0.05)。结论姜黄素可保护心肌组织,其机制可能是通过提高心肌组织的抗氧化能力,减轻氧化应激损伤,抑制或降低炎症因子的释放。
Objective To investigate the effect of curcumin on expressions of serum myocardial enzymes,oxidative stress enzymes and inflammatory cytokines in mice with sleep deprivation. Methods Fifty male C57 mice were randomly divided into normal sleep group,sleep deprivation(SD) model group as well as low,moderate and high dose curcumin groups. The model was established by sleep deprivation with improved multi-platform water environment method for 72 h. The mice in low,moderate and high dose curcumin groups were treated with curcumin at dosages of 50,100 and150 mg/kg by gavage respectively,while those in normal sleep a nd SD model groups with an equal volume of normal saline,once a day for 8 weeks. Serum lactate dehydrogenase(LDH),creatine kinase(CK),alpha-hydroxybutyrate dehydrogenase(α-HBDH) and aspartate aminotransferase(AST) contents were determined by BS-220 fully automatic biochemical analyzer,while the superoxide dismutase(SOD) activity in myocardial tissue by xanthine oxidase method,the glutathione peroxidase(GSH-Px) activity by colorimetry,the malondialdehyde(MDA) content by thibabituric acid(thiosulfuric acid barbitone,TBA)method,and the interleukin-1β(IL-1β),interleukin 6(IL-6)and tumor necrosis factor-α(TNF-α)contents in myocardial tissue by ELISA. Results Compared with those in SD model group,the contents of LDH,CK,α-HBDH and AST in sera of mice in low,moderate and high dose curcumin groups decreased(P〈0. 000 1),while the activities of SOD and GSH-Px in myocardial tissue increased gradually(P〈0. 05),the content of MDA decreased gradually(P〈0. 05),and the contents of IL-1β,IL-6 and TNF-α decreased gradually(P〈0. 05). Conclusion Curcumin may protect myocardium,of which the mechanism may be reducing oxidative stress injury and inhibiting or reducing the release of inflammatory cytokines through improving the antioxidant capacity of myocardial tissue.
