柴达木黑果枸杞原花青素对小鼠衰老皮肤抗氧化作用及炎症因子的影响 被引量:5

Anti-skin aging effects of proanthocyanidins in Lycium ruthenicum from Chaidamu by Antioxidant and inflammatory factors in mice

摘要 目的探讨柴达木黑果枸杞原花青素对皮下注射5%D-半乳糖(D-gal)联合紫外线(长波紫外线UVA+中波紫外线UVB)照射后小鼠皮肤的抗氧化作用及炎症因子的影响。方法将50只昆明小鼠随机分为5组(正常组,模型组,维生素E组,原花青素低剂量组、高剂量组),连续造模40天。造模完成后,取小鼠颈背部皮肤制备10%的组织匀浆,取上清液,用生化法测超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)和过氧化氢酶(CAT)活力;用ELISA法测白细胞介素IL-1、IL-6含量。结果与正常组比较,模型组SOD活力和GSH-PX、CAT含量降低,IL-1、IL-6含量增加显著(P<0.05);与模型组相比,维生素E组、原花青素低、高剂量组的SOD活力和GSH-PX、CAT含量不同程度增加,IL-1、IL-6含量降低(P<0.05);原花青素低、高剂量组与维生素E组比较差异均有统计学意义(P<0.05)。结论柴达木黑果枸杞原花青素能够减轻紫外线照射联合皮下注射D-半乳糖对小鼠皮肤的氧化损伤;减少IL-1、IL-6的分泌,减轻炎症反应,延缓皮肤衰老,且效果优于同等剂量的维生素E。 Objective To investigate the anti-aging skin effect of the proanthocyanidins in Lycium ruthenicum from Chaidamu and Vitamin E of 5% D-galactose combined with ultraviolet UV(UVA+UVB) radiation.Method Fif- ty healthy female rats were randomly divided into 5 groups( normal group, aging model group, control group ,low con- centration of proanthocyanidins in Lycium ruthenicum group, high concentration of proanthocyanidins in Lycium ru- thenicum group), continuously modeling for 40 days.After finishing the molding, take off the skin around the back and neck,enzyme biochemical method measure SOD activity, GSH-PX content, CAT activity;and the content of IL-1 and IL-2 was detected by ELISA.Result Comparison with the normal group,the activity of SOD and the con- tent of GSH-PX and CAT in model group were reduced,the content of IL-1 and IL-2 were increased(P〈0.05 ) ;Contrast with the model group,the activity of SOD and the contents of GSH-PX and CAT in the control group and low concentration of proanthocyanidins in Lycium ruthenicum group and high concentration of proanthocyanidins in Lycium ruthenicum group were increased ,the contents of IL-1 and IL-2 were reduced (P〈0.05) ;And comparing the low concentration of proanthocyanidins in Lycium ruthenicum group or high concentration of proanthocyanidins in Lycium ruthenicum group with the control group, the difference between the two groups was statistically signifi- cant(P〈0.05) .Conclusion The proanthocyanidins in Lycium ruthenicum from Chaidamu can inhibit the oxidative damage of D-galactose combined with ultraviolet ( UVA+UVB ) radiation, and down-regulation of the expression lev- el of IL-1 and IL-2, alleviate the inflammation, delay senescence of skin in a certain degree, and the effect is better than vitamin E.
