摘要
目的建立和评价大鼠门静脉血栓模型。方法将SD大鼠随机分为假手术组和模型组,模型组通过门静脉暂时结扎减慢血流,联合钳夹破坏血管内膜的方法建立大鼠门静脉血栓模型,而假手术组仅分离门静脉;通过B超检测术后24h门静脉血栓的形成;通过HE染色观察门静脉的形态学变化。结果 B超检查发现模型组的门静脉于术后24h形成稳定血栓,而假手术组门静脉未检测到血栓形成;HE染色检测发现模型组门静脉存在大量混合血栓,血管内膜损伤、脱落及炎细胞浸润;假手术组门静脉血管内膜完整,未见血栓形成。结论采用本文方法,24小时可成功建立大鼠门静脉血栓模型,操作简易、模型稳定、符合临床特点。
Objective This study aimed to establish and evaluate a rat model with portal vein thrombosis(PVT).Methods SD rats were randomly divided into model group and sham operation group.The PVT model was established by ligating portal vein briefly to slow blood flow and clamping vessel wall repeatedly to damage endangium,while portal vein was only dissociated in sham operation group.The portal vein thrombosis was detected by using Bmode ultrasonic examination at 24 h.HE staining was used to detect the morphology of the portal vein.Results Bmode ultrasonic examination showed that stable thrombosis was formed in the portal veins of model group at 24 hafter the operation,while no thrombosis was observed in the portal veins of sham operation group.HE staining showed that the portal vein was filled with mixed thrombus.Its endangium was damaged and shed with the infiltration of a lot of inflammatory cells.In sham operation group,the endangium of the portal veins was intact,and no thrombus was formed.Conclusion The rat PVT model can be successfully established within 24 hours by this method,which is simple,stable and in accordance with clinical characteristics.
