清鼻丸对鼻—鼻窦炎家兔鼻黏膜组织Caspase-3和p38MAPK的影响 被引量:2

摘要 目的探讨清鼻丸对鼻—鼻窦炎家兔鼻黏膜组织Caspase-3和p38丝裂原活化蛋白激酶(p38MAPK)的影响。方法将48只实验家兔随机分为正常组、假手术组、模型组、清鼻丸组,每组12只。采用窦口不完全堵塞加金黄色葡萄球菌诱导的兔慢性鼻—鼻窦炎模型。正常组不造模,不灌胃,常规喂养。假手术组造模,但窦腔内不填塞棉絮,不注入金黄色葡萄球菌悬浊液,模型组造模,两组自造模后第43天开始,每日以等量生理盐水灌胃。清鼻丸组自造模后第43天开始,给予清鼻丸14 g生药/(kg·d)灌胃。免疫组化法检测兔鼻窦黏膜组织Caspase-3蛋白表达,反转录-聚合酶链式反应(RT-PCR)检测p38MAPK mRNA表达。结果与正常组比较,模型组Caspase-3阳性细胞表达数目相对较多,p38MAPK mRNA表达显著增加,差异均具有统计学意义(P<0.01)。与模型组比较,清鼻丸组Caspase-3阳性细胞和p38MAPK mRNA表达显著下调,差异均具有统计学意义(P<0.01)。结论清鼻丸具有改善鼻—鼻窦炎病理作用,其作用机制可能与其抑制Caspase-3和p38MAPK表达有关。 Objective To explore the effects of qingbi pills on mucosal Caspase 3 and p38 MAPK in the rabbits with nasosinusitis. Methods A total 48 experimental rabbits were randomized into a normal group,a sham-operation group,a model group and a qingbi pills group,12 rabbits in each one. The model of chronic nasosinusitis was induced by the incomplete sinus block and staphylococcus aureus. In the normal group,no modeling and lavage were provided. The rabbits were fed normally. In the sham-operation group,the model was prepared,but without sinus block and infusion of staphylococcus aureus suspension. In the model group,the model was prepared. In these two groups,from the 43 rdday of modeling,the lavage with the physical saline of the same amount was used every day. In the qingbi pills group,starting from the 43 rdday of modeling,the lavage was given with the raw herbs of qingbi pills,14 g/(kg·d). The IHC method was used to determine the protein expression of Caspase-3 and RT-PCR was to determine the expression of p38 MAPK mRNA.Results Compared with the normal group,the expression of Caspase-3 positive cell was relatively more in the model group and the expression of p38 MAPK mRNA was increased significantly,indicating the significant differences(P〈0. 01). Compared with the model group,the expressions of Caspase-3 positive cell and p38 MAPK mRNA were down-regulated significantly in the qingbi pills group(P〈0. 01). Conclusion Qingbi pills relieve the pathological effects on nasosinusitis and the mechanism is probably related to the inhibition of the expressions of Caspase-3 positive cell and p38 MAPK mRNA.
