摘要
目的研究Kiss1对鼻咽癌转移能力的影响及其可能机制。方法首先通过裸鼠鼻咽癌肺转移模型观察移植瘤与转移瘤中Kiss1的表达差异,继而通过transwell实验在细胞水平检测Kiss1细胞过表达后的侵袭转移能力的变化,通过qRT-PCR和western-blot检测MMPs家族的相关分子表达变化。结果裸鼠实验发现Kiss1在肺转移瘤中的表达较移植瘤低,细胞实验发现过表达Kiss1后鼻咽癌细胞的转移能力降低,qRT-PCR和western-blot检测发现过表达Kiss1后MMP9表达降低。结论通过动物实验论证Kiss1基因可抑制鼻咽癌细胞的侵袭转移。推测MMP9是Kiss1的下游靶基因,在Kiss1基因抑制肿瘤转移过程中发挥着重要作用。
Objective To study the influence of Kiss1 on nasopharyngeal carcinoma metastasis ability and its possible mechanism.Methods The expression of Kiss1 in transplanted tumor and metastatic tumor were observed in nude mice model of nasopharyngeal carcinoma. The invasion and metastasis ability of nasopharyngeal carcinoma cells was detected by Transwell assay after up-regulated the expression of Kiss1. qRT-PCR and western-blot were used to detect the expression of MMPs after up-regulated the expression of Kiss1.Results The expression of Kiss1 in lung metastasis was lower than that of the transplanted tumor in nude mice. After up-regulated the expression of Kiss1,the invasion activity of nasopharyngeal carcinoma cells decreased,as well as the expression of MMP9. Conclusions Animal experiments showed that Kiss1 gene can inhibit invasion and metastasis of nasopharyngeal carcinoma cells. It is speculated that MMP9 is the downstream target gene of Kiss1 and plays an important role in the inhibition of tumor metastasis by Kiss1 gene.
