摘要 目的探讨Rab1A蛋白在胃腺癌(Gastric adenocarcinoma,GAC)组织中表达情况以及其表达水平与患者预后的影响。方法组织芯片技术、免疫组化En Vision法检测Rab1A在平均随访5年的300例胃腺癌组织中表达水平。通过SPSS 17.0软件分析胃腺癌组织中Rab1A蛋白的表达情况与胃腺癌患者临床病理之间的关系,以及Rab1A蛋白表达水平对患者预后的影响。结果胃腺癌组织中Rab1A蛋白高表达率明显高于癌旁非癌胃组织(82.26%vs 31.43%,χ2=12.825,P=0.0016);在高-中分化(23.2%)、T1-3分期(31.5%)及临床分期Ⅰ~Ⅱ期(48.3%)胃腺癌组织中Rab1A蛋白高表达较低(P<0.05);生存(Kaplan-Meier法)分析显示,Rab1A蛋白表达低组比Rab1A蛋白表达高组的生存期明显延长,无进展生存期(Progression-free surial,PFS)及总生存期(Overall survival,OS)均延长,差异有统计学意义(P<0.05)。结论 Rab1A蛋白高表达对胃腺癌的发生发展具有促进作用,可作为胃腺癌患者预后评估的重要指标及靶向治疗的潜在靶标。 Objective To investigate the expression of Rab1 A protein in gastric adenocarcinoma tissue and its prognostic value. Methods Tissue microarray technology and immunohistochemically En Vision method were used to detect the expression of Rab1 A in 300 cases of gastric adenocarcinoma tissue. The average follow up time of patients was about 5 years. Correlations between expressions of Rab1 A protein in glioma and clinical features and prognosis were analyzed by the software SPSS 17. 0. Results The high expression rate of Rab1 A protein was significantly higher in the gastric adenocarcinoma tissues than the adjacent normal esophageal mucosa( 82.26% vs 31. 43%,χ2= 12. 825,P = 0. 0016). The expression of Rab1 A protein was low in the gastric adenocarcinoma tissues of high and moderate differentiation( 23.2%),T1-T3 stage( 31.5%) and clinical stages I to II( 48.3%)( P 〈 0.05). Survival analysis( Kaplan Meier method) suggested that progression-free survival( PFS) and overall survival( OS) of the patients with low Rab1 A protein expression were longer than those with high expression( P 〈 0.05). Conclusions High Rab1 A protein expression may promote the incidence of gastric adenocarcinoma. It could be used as an important indicator of prognosis and as a potential target to develop targeted therapy.
