摘要
建立气相色谱法测定羊肚菌(干品)中脂肪酸的方法。以石油醚(30~60℃)为提取溶剂,利用索氏提取器提取羊肚菌(干品)中的脂肪酸,旋转蒸干石油醚,加4 mL正己烷,0.1 mL氢氧化钾-甲醇溶液,涡旋1 min,以转速4 000 r/min离心5 min,取上清液,甲酯化后得样品溶液,进样分析。结果表明,饱和脂肪酸(SFA)在4批次样品中含量均较低,分别为1.860%,1.745%,0.619%,0.625%;单不饱和脂肪酸(MUFA)在4批次样品中含量均较高,分别为76.834%,71.851%,72.527%,76.802%;多不饱和脂肪酸(PUFA)含量也较高,含量分别为22.528%,24.946%,26.870%,22.423%。
To establish a GC method for the determination of fatty acids in Morchella. Using petroleum ether (30~60℃)as extraction solvent, soxhlet extractor was used to extract fatty acids from Morchella esculentum. Rotary evaporated petroleum ether was added. 4 mL of n-hexane and 0.1 mL of potassium hydroxide-methanol solution were added and vortexed for 1 min, centrifuge 5 min, 4 000 r/min, take the supernatant, methyl esterify the sample solution, and analyzed the sample. The re-sults showed that the content of saturated fatty acids (SFA) was low in four batches of samples, which were 1.860% , 1.745%, 0.619%, and 0.625%, respectively. Monounsaturated fatty acids (MUFA) were higher in four batches of samples. They were 76.834%, 71.851%, 72.527%, 76.802% respectively. The polyunsaturated fatty acid (PUFA) content was also higher, with the contents being 22.528%, 24.946%, 26.870%, and 22.423%, respectively.
