补肾健骨汤干预PMOP小鼠BMSCs成骨分化机制的实验研究

Study on PMOP Mice's BMSCs Osteogenesis Differentiation Mechanism Intervened by Bushen Jiangu Decoction

目的:观察去除卵巢小鼠股骨中骨髓间充质干细胞的成骨分化机制来探究补肾健骨汤治疗绝经后骨质疏松症的临床疗效。方法:本实验研究采取去除小鼠双侧卵巢的方法复制绝经后骨质疏松症模型,将实验小鼠随机分为假手术组、补肾健骨汤组和生理盐水组,手术后一周开始给药干预,2次/天,连续给药15天后通过碱性磷酸酶染色和茜素红染色来检测三组小鼠骨髓间充质干细胞的成骨分化机制。结果:补肾健骨汤组的碱性磷酸酶检测水平明显高于生理盐水组(P <0. 05),补肾健骨汤组的茜素红染色检测水平也明显高于生理盐水组(P <0. 05)。结论:运用补肾健骨汤来治疗绝经后骨质疏松症模型小鼠取得了良好的效果,因此,认为补肾健骨汤中的药物成分对骨髓间充质干细胞的增值和分化具有推动作用,它能够促进骨骼的修复和生成,对治疗绝经后骨质疏松症具有良好的效果。

Objective ： To study the clinical effects of Bushen Jiangu Decoction which can be effective at treating postmenopausal osteoporosis （PMOP） by observing the change of the Bone Marrow Mesenchymal Stem Cells （BMSCs） osteogenesis differentiation in femurs of the mice, whose ovaries were removed. Methods： The study duplicated PMOP models by removing the bilateral ovaries of mice and divided the mice into three groups ： the sham operation group, Bushen Jiangu Decoction group, and physiological saline group. One week after operation, they were intervened with drugs twice a day, after fifteen days, by alkaline phosphatase （ALP） and Alizarin Red to detect mice＇ s BMSCs differentiation mechanism in three groups. Result： ALP of the Bushen Jiangu Decoction group was obviously higher than that of physiological saline group （P 〈 0.05 ） , also Alizarin Red of the Bushen Jiangu Decoction group was obviously higher than that of physiological saline group （ P 〈 0.05 ）. Conclusion ： Bushen Jiangu Decoction has an effective treatment at PMOP model mice, so its action mechanism promotes the accretion of BMSCs in the femur of mice to a certain degree and the formation of bone tissue, so as to achieve the purpose of treating PMOP.
