摘要
目的实验对象为THP-1巨噬细胞,将巨噬细泡沫化,在最佳时间(16 h内),浓度不同的丹酚酸B,对泡沫细胞内胆固醇代谢和ATP结合盒转运体G1 (ABCG1) mRNA的表达影响。方法将THP-1细胞在50 nmol/L佛波酯(PMA)诱导下,分化成巨噬细胞,应用氧化型低密度脂蛋白诱导巨噬细胞泡沫化,建立巨噬细胞泡沫细胞模型;将不同浓度丹酚酸B在最优时间16 h内加入泡沫化巨噬细胞,细胞内的脂类含量采用油红O染色法测定,细胞内胆固醇酯(CE)占总胆固醇(TC)含量比值采用高效液相色谱检测法进行分析研究,荧光实时定量RT-PCR检测法测定ABCG1的mRNA表达。结果在100μmol/L丹酚酸B实验组中,ABCG1 mRNA的表达明显增加(为对照组的2. 12倍,P <0. 05)。结论丹酚酸B可限制THP-1巨噬细胞泡沫化,这与丹酚酸B增加巨噬细胞中ABCG1的表达,从而促进巨泡沫细胞中胆固醇的流出有关。
Objective To foam macrophage THP-1 which was the research object and observe the effects of different concentrations of Danphenolic acid B on cholesterol metabolism in foam cells and the expression of the ATP binding box transporter G1 ( ABCG1 ) mRNA during the optimal time. Methods THP-1 cells were induced by 50 nmol/L phorbol (PMA) to differentiate into macrophages and the macrophage foam was induced by oxidized low-density lipoprotein to establish the macrophage foam cell model. Foam macrophages were added to different concentrations of salvianolic acid B within the optimal time of 16 h. The intracellular lipid content was determined by oil red O staining, the ratio of intracellular cholesterol ester (CE) to total cholesterol (TC) was analyzed by HPLC, and the mRNA expression of ABCG1 was determined by fluorescence real-time quantitative RT-PCR. Results The expression of ABCG1 mRNA was significantly increased in the 100 μmol/L salvianolic acid B experimental group (2. 12 times of that in the control group, P 〈0. 05 ). Conclusion Salvianolic acid B can limit THP-1 macrophage foaming, which is related to the increased expression of ABCG1 in macrophages by salvianolic acid B, thereby promoting cholesterol efflux in macro-foam cells.
