DC细胞疫苗干预肝癌Treg细胞及与IDO、IFN-γ等细胞因子相关性 被引量:4

摘要 目的分析树突状细胞(DC)细胞疫苗干预对肝癌调节性T细胞(Treg)的影响及其与细胞因子吲哚胺2,3-双加氧酶(IDO)、干扰素(IFN)-γ、白介素(IL)-10、IL-12、转化生长因子(TGF)-β的相关性。方法选取2015年3月至2018年1月期间我院90例经DC细胞疫苗干预的肝癌患者为病例组,另选取同期入院体检的健康志愿者50例作为对照组,对照组体检时,病例组于DC细胞疫苗干预前、干预后14d采血,检测外周血T淋巴亚群及细胞因子IDO、IL-10、IL-12、IFN-γ、TGF-β水平,并评价Treg细胞与上述细胞因子的关系。结果与对照组相比,病例组干预前外周血CD3^+CD4^+、CD3^+CD8^+、CD3^+CD56^+水平明显低,Treg细胞及血细胞因子IDO、IL-10、IL-12、IFN-γ、TGF-β水平明高,干预后CD3^+CD4^+仍明显低,Treg细胞及血细胞因子IDO、IL-10、IL-12、IFN-γ、TGF-β水平仍明显高,差异均有统计学意义(P<0.05);但与干预前比较,病例组干预后CD3^+CD4^+、CD3^+CD8^+、CD3^+CD56^+、Treg细胞水平均显著改善,IDO、IL-10、IL-12、IFN-γ、TGF-β水平均显著下降(P<0.05)。DC细胞疫苗干预后,肝癌患者外周血Treg细胞与细胞因子IDO、IL-10、IL-12、IFN-γ、TGF-β水平正相关(P<0.05)。结论 DC细胞疫苗干预可调节肝癌患者外周血Treg细胞及细胞因子IDO、IL-10、IL-12、IFN-γ、TGF-β含量,且Treg细胞含量变化与IDO、IFN-γ等细胞因子具有显著相关性。 Objective To analyze the effect of dendritic cell (DC) intervention on regulatory T cells (Treg) in hepatocellular carcinoma and the correlation with indoleamine 2,3-dioxygenase (IDO), interferon (IFN) γ, interleukin (IL) -10, IL-12 and transforming growth factor (TGF) - β. Methods 90 patients with hepatocellular carcinoma who were treated with DC vaccine during March 2015 January 2018 were selected as the case group, and another 50 healthy volunteers who underwent physical examination in the hospital during the same period were selected as the control group. The blood samples of case group were collected before DC vaccine intervention and at 14d after the intervention to detect levels of T lymphocyte subsets and cytokines IDO, IL-10, IL-12, IFN-γ and TGF-β in peripheral blood. The relationship between Treg cells and the above cytokines was evaluated. Results Compared with the control group, the levels of peripheral blood CD3 +CD4 +, CD3 +CD8 + and CD3 +CD56 + were significantly lower, while levels of Treg cells and blood cytokines IDO, IL-10, IL-12, IFN-γ and TGF-β were higher in the case group before the intervention. After the intervention, the level of CD3 +CD4 + was lower, while levels of Treg cells and blood cytokines IDO, IL-10, IL-12, IFN-γ and TGF-β were significantly higher in the case group than in the control group ( P 〈0.05). After the intervention, levels of CD3 +CD4 +, CD3 +CD8 +, CD3 +CD56 + and Treg cells in the case group were significantly improved, and levels of IDO, IL-10, IL-12, IFN-γ and TGF-βdecreased significantly ( P 〈0.05). After the intervention, Treg cells in peripheral blood of patients with hepatocellular carcinoma were positively correlated with the levels of cytokines IDO, IL-10, IL-12, IFN-γ and TGF-β ( P 〈0.05). Conclusion DC vaccine intervention can regulate the levels of Treg cells and cytokines IDO, IL-10, IL-12, IFN-γ and TGF-β in peripheral blood of patients with hepatocellular carcinoma, and the change of Treg cell content is significantly correlated with cytokines such as IDO and IFN-γ.
