摘要
目的:观察"嗅三针"对阿尔茨海默病(AD)小鼠学习和海马IL1-β和IL-6表达的影响,以阐明其基于嗅觉通路治疗AD的作用机制。方法:采用APP/PS1转基因小鼠作为AD小鼠模型,小鼠嗅觉通路阻断模型采用嗅神经切断术建立,将40只AD小鼠随机分为模型对照组、嗅神经切断嗅三针组、嗅三针组、盐酸多奈哌齐组,每组10只,另取10只同窝的阴性小鼠作为正常对照组。通过"嗅三针"对印堂穴和双侧迎香穴行电针刺激,每次10分钟,以3mg/kg/d的剂量对盐酸多奈哌齐组AD小鼠进行灌胃,每周干预5次,间歇2天,共干预4周。小鼠学习能力采用Morris水迷宫进行监测,应用Western blot技术检测海马IL1-β和IL-6表达水平。结果:嗅三针刺激和盐酸多奈哌齐灌胃能够显著缩短AD小鼠平均逃避潜伏期,并能显著降低海马IL1-β和IL-6的表达,与AD模型组比较,差异具有显著性(P<0. 05);嗅神经切断嗅三针组对AD小鼠平均逃避潜伏期以及海马IL1-β和IL-6的表达均无明显影响,与AD模型组比较,差异无显著性(P>0. 05),嗅三针组与盐酸多奈哌齐组比较无显著性差异(P>0. 05)。结论:"嗅三针"具有改善AD小鼠学习能力,降低海马水平IL1-β和IL-6表达的作用,嗅觉通路的完整性可能是其发挥干预效应的核心机制。
Objecticve: To observe the intervening effect of"Xiusanzhen"Electroacupuncture( EA) on learning and memory ability and IL1-β,IL-6 expression in hippocampus of mice with Alzheimer's disease( AD),in order to clarify its therapeutic mechanism based on the olfactory nerve pathway in alleviating AD. Methods: APP/PS1 transgenic mice were used as AD mice models,mice model with olfactory nerve pathway block were established by transecting olfactory nerve,40 AD mice were randomly devided into 4 groups: AD model group,"Xiusanzhen"EA with olfactory nerve transected group( ONT+EA),"Xiusanzhen"EA group,Donepezil HCl group,with 10 mice respectively in each group,then took 10 negative mice as normal controlled group.Electroacupuncture intervention of"Xiusanzhen"was applied to stimulate bilateral LI 20 and EX-HN3 for 10 min per time,intragastric administration of donepezil HCI in AD mice was performed at the dose of 3 mg/kg/d,once daily intervention( except weekends) for 4 weeks in all. Morris water maze was applied to test learning ability of mice. RT-PCR and agarose gel electrophoresis were applied to test IL1-β and IL-6 expression in hippocampus of mice. Results: Compared with AD model group, "Xiusanzhen"EA group Donepezil HCl group could shorter mean escape latency,meanwhile,reduce IL1-β and IL-6 expression in hippocampus of AD mice,there were significant significant differences between the two groups( P〈0. 05); There were no significant differences were found between AD model and ONT + EA groups in influencing mean escape latency and IL1-β,IL-6 expression in hippocampus of AD mice( P〉0. 05),there were no significant difference between "Xiusanzhen"EA group and donepezil HCl group( P〉0. 05). Conclusion: "Xiusanzhen"EA has the effect in ameliorating learning ability and reducing the IL1-β and IL-6 expression in hippocampus of mice with AD,the integrity of the olfactory pathway is the core mechanism of the intervention effect.
