摘要
为了开展猪渗出性皮炎的噬菌体治疗,利用双层平板法进行噬菌体的分离、纯化及电镜观察,并对噬菌体的最佳感染复数、一步生长曲线、温度及pH值稳定性以及体外裂解能力等进行检测。结果显示,分离到1株烈性噬菌体vB-StaMJTB,为肌尾噬菌体。最佳感染复数为0. 000 1,噬菌体的潜伏期80 min,裂解期为220 min,裂解量为77;在pH值为5~9以及低于50℃条件下保持很好的稳定性;该噬菌体对猪源葡萄球菌的裂解率为71. 93%,且在体外裂解试验中12 h未出现抗性菌。
In order to provide phage therapy for swine exudative epidermitis, phage of Staphlyococeus from swine was isolated and purified by using double-plate agar method and observed by electron microscope. The optimal multiplicity of infection (MOI) , one-step growth curve, temperature and pH stability, and external lysis ability were als0 determined. The results showed that a viru- lent Myoviridae bacteriophage vB-StaM-JTB was isolated. The optimal MOI was 0. 0001, and the latent period was 80 min. The lysis period was 220 min and the burst size was 77. The phage could keep its good stability in the pH range of 5 to 9 and at the tempera- ture lower than 50 ℃. 71.93% of Staphlycoceus strains from swine could be lyzed by the phage and no resistant bacterial strains ap- peared within 12 h.
