摘要
目的建立HPLC-ELSD法同时测定十味鹅黄颗粒(黄芪、辛夷、柴胡等)中3种成分的含有量。方法该药物10%氨水-水饱和正丁醇-甲醇提取液的分析采用ZORBAX Eclipse XDB-C_(18)色谱柱(4. 6 mm×250 mm,5μm);流动相乙腈-水,梯度洗脱;体积流量1. 0 mL/min;柱温30℃。结果黄芪甲苷、柴胡皂苷A、木兰脂素分别在0. 081 6~0. 816 mg/mL (r=0. 995 0)、0. 019 1~0. 191 mg/mL (r=0. 993 6)、0. 051 8~0. 518 mg/mL (r=0. 994 3)范围内线性关系良好,平均加样回收率分别为101. 5%、100. 8%、101. 7%,RSD分别为0. 48%、0. 57%、0. 34%。结论该法简便可靠,重复性好,可用于十味鹅黄颗粒的质量控制。
AIM To establish an HPLC-ELSD method for the simultaneous content determination of thine con- stituents in Shiwei Ehuang Granules ( Astragali Radix, Magnoliae Flos, Bupleuri Radix, etc. ). METHODS The analysis of 10% aqueous ammonia- (water-saturated n-butanol) -methanol extract of this drug was performed on a 30℃ thermostatic ZORBAX Eclipse XDB-C18 scolunm (4.6 mm × 250 ram, 5 μm), with the mobile phase comprising of acetonitrile-water flowing at 1.0 mL/min in a gradient elution manner. RESULTS Astragaloside A, saiko- saponin A and magnolin showed good linear relationships within the ranges of 0. 081 6 - 0. 816 mg/mL ( r = 0. 995 0), 0. 019 1 -0. 191 mg/mL (r =0. 993 6) and 0. 051 8 -0. 518 mg/mL (r =0. 994 3), whose average recoveries were 101.5% , 100. 8% and 101.7% with the RSDs of 0.48% CONCLUSION This simple, reliable and reproducible method can be Ehuang Granules. 0.57% and 0.34% , respectively used for the quality control of Shiwei
