摘要
目的调查20个STR基因座(D1S1656、D5S818、D8S1179、D7S820、CSF1PO、D2S1338、D3S1358、vWA、D21S11、D16S539、D12S391、PentaE、FGA、TPOX、TH01、D6S1043、D19S433、D18S51、D13S317、Penta D)在黑龙江省绥化地区汉族人群中的遗传多态性。方法对2627名汉族无关个体样本,采用PowerPlex?21试剂盒以9700型扩增仪作复合扩增, ABI 3130xL型遗传分析仪基因分型。所得STR分型数据使用自主研发的法医物证基因分型提取软件采集,并以Modified-Powerstats和Cervus3.0分析。结果 20个STR基因座共检出277个等位基因和1276种基因型;除PentaE、D16S539、D5S818、D12S391四个基因座外,其余基因座的基因型频率分布均符合Hardy-Weinberg平衡(P> 0.05),杂合度(He)值在0.619~0.873之间,个体识别能力(DP)值在0.791~0.988之间,二联体非父排除率(PE2)值在0.359~0.839之间,三联体非父排除率(PE3)值在0.527~0.957之间。结论 16个STR基因座在所调查的人群中具有较高的识别能力。获得的数据可为黑龙江绥化地区汉族人群基础研究和法医学应用研究提供科学的基础数据。
Objective To investigate the genetic polymorphism of 20 autosomal STR loci (D1S1656, D5S818, D8S1179, D7S820, CSF1PO, D2S1338, D3S1358, vWA, D21S11, D16S539, D12S391, PentaE, FGA, TPOX, TH01, D6S1043, D19S433, D18S51, D13S317, Penta D) among Han population in Suihua area of Heilongjiang Province. Methods From 2,627 unrelated Han-ethnic individuals in the designated area, the 20 STR loci indicated above were selected and produced with multiplex amplification by PowerPlex 21 Kit on 9700 PCR apparatus, with their genotypes being obtained by genetic analyzer (ABI 3130XL). The STR data were extracted by self-developed software and analyzed with the Modified-Powerstats and Cervus3.0. Results From the tested individuals, there were found 244 alleles and 1276 genotypes of the 20 STR loci. Except for the four loci of PentaE, D16S539, D5S818 and D12S391, the other 16 loci showed their genotype frequency distributions in accordance with Hardy-Weinberg equilibrium (P〉0.05), He value between 0.619~0.873, DP value amid 0.791~0.988, PE2 value within 0.359~0.839, and PE3 value spanning across 0.527~0.957. Conclusion Of the 20 STR loci tested, 16 STR loci showed higher recognition ability in the investigated population. These data can provide basis for both research and forensic application of Han-ethnic population in Suihua area of Heilongjiang Province.
