摘要
目的建立一种可同时检测降香挥发油及水溶极性成分的方法,实现精制冠心片中降香药味的质量控制。方法采用硅胶G板为薄层板,以石油醚(60~90℃)-乙酸乙酯(8∶3)为展开剂,同时检测降香中挥发油和水溶极性成分;采用气相色谱法,HP-5毛细管柱(30 m×0. 32 mm,0. 25μm),FID检测器,以反式橙花叔醇为检测指标,对降香挥发油的检测结果作进一步确证。结果采用薄层色谱法可实现一个色谱条件同时检测降香挥发油和水溶极性成分,方法比移(Rf)值适中,各组分斑点分离度良好;气相色谱法中反式橙花叔醇与其他组分分离度良好,检出限为0. 105μg/g,检测灵敏度高,能快速、有效地对降香挥发油的检测结果进行进一步确证。结论该试验中建立的方法可实现对精制冠心片中降香药味的质量控制,为进一步规范降香的投料和生产工艺提供了依据。
Objective To establish a method for simultaneous detection of volatile oil and water-soluble component of Lignum Dalbergiae Odoriferae in Jingzhi Guanxin Tablets,in order to control the quality of Lignum Dalbergiae Odoriferae in Jingzhi Guanxin Tablets. Methods Silica gel G plate was taken as thin layer plate,petroleum ether( 60-90 ℃)-ethyl acetate( 8 ∶ 3) was taken as developer,volatile oil and water-soluble component of Lignum Dalbergiae Odoriferae were identified simultaneously. The detection results of volatile oil of Lignum Dalbergiae Odoriferae were further verified by GC with HP-5 capillary column( 30 m × 0. 32 mm,0. 25 μm)and FID detector with trans-nerolidol as the detection index. Results Volatile oil and water-soluble component of Lignum Dalbergiae Odoriferae could be detected simultaneously under a same TLC condition with moderate Rf value and good separation degree. The trans-nerolidol was separated absolutely from other component by GC method,LOD was 0. 105 μg/g,and the GC method has high sensitivity and could be used to verify the TLC detection results of volatile oil of Lignum Dalbergiae Odoriferae rapidly and effectively.Conclusion The established method can realize the quality control of Lignum Dalbergiae Odoriferae in Jingzhi Guanxin Tablets and provide a basis for further regulation of the feeding and production process of Lignum Dalbergiae Odoriferae.
