摘要
目的:研究玉米紫色植株色素(maize purple plant pigment,MPPP)预干预对过氧化氢(H2O2)诱导的MC3T3-E1成骨细胞氧化损伤的影响。方法:采用MTT法观察浓度为10-10~10-3mol/L的H2O2对成骨细胞增殖能力的影响,并选择最佳浓度建立MC3T3-E1成骨细胞氧化损伤模型(H2O2处理组);同时选择MPPP促进成骨细胞增殖的适宜浓度,采用化学比色法观察H2O2分别对低(10-7mol/L)、中(10-6mol/L)、高(10-5mol/L)浓度的MPPP预干预24、48和72 h后的成骨细胞活力及MPPP预干预72 h后各组细胞总抗氧化力(T-AOC)的影响。结果:10-7~10-3mol/L的H2O2均可抑制成骨细胞增殖(P<0.01)。不同浓度MPPP在不同预干预时间条件下,H2O2(5×10-4mol/L)处理后各组细胞OD值均显著低于对照组(P<0.05或0.01),仅预干预72 h的高浓度MPPP组细胞OD值高于H2O2处理组(P<0.05);MPPP预干预72 h后,与对照组比较,H2O2处理组和低、中、高浓度MPPP组成骨细胞T-AOC均显著降低(P<0.01),但高浓度MPPP组成骨细胞T-AOC水平明显高于H2O2处理组(P<0.01)。结论:MPPP预干预可抑制H2O2诱导的MC3T3-E1成骨细胞的氧化损伤并提升成骨细胞T-AOC。
Objective: To investigate the effects of different concentrations of maize purple plant pigment (MPPP) pre-intervention on oxidative damage of osteoblast MC3T3-E1 treated with hydrogen peroxide (H2O2). Methods: MC3T3-E1 osteoblasts were treated with different concentration of H2O2 (10^-10~10^-3mol/L) for 1 h, and the optimum concentration was selected to establish the cell oxidative damage model. After that, MC3T3-E1 osteoblasts were pretreated with different doses of MPPP for 24, 48 and 72 h respectively and stimulated with H2O2 (5×10^-4mol/L)for 1 h. MTF assay and chemistry eolorimetry were used to determine the proliferation and total antioxidant capacity (T-AOC) of MC3T3-E1 cell exposed to H2O2 (5×10^-4mol/L)by MPPP pre-intervention respectively. Results : The proliferation of MC3T3-E1 osteoblasts was significantly inhibited after H2O2 treatment (P 〈 0.05), while after 72 h pre-intervention, promoting proliferation of MC3T3-E1 cells treated with MPPP (10^-5 mol/L) was found (P〈 0.05). Chemistry eolorimetry showed that MPPP (10^-5 mol/L) elevated T-AOC level of MC3T3-E1 cells treated with H2O2 after 72 h pre-intervention of MPPP (P 〈 0.01). Conclusion: MPPP pre-intervention can promote proliferation and elevate T-AOC level of MC3T3-E1 osteoblasts treated with H2O2.
