色素上皮衍生因子及受体在皮脂腺SZ95细胞的表达

摘要 目的探讨色素上皮衍生因子(PEDF)及受体在皮脂腺细胞系SZ95细胞的表达。方法采用RT-PCR法检测SZ95细胞中PEDF及受体的mRNA表达,蛋白免疫印迹法检测PEDF及受体蛋白的表达,同时免疫荧光法定位其在SZ95细胞中的表达情况。使用不同浓度双氢睾酮(DHT)作为刺激因子作用于SZ95细胞,实时定量PCR (qPCR)和免疫印迹法检测PEDF mRNA和蛋白水平变化。结果 RT-PCR发现PEDF及受体在SZ95细胞中均有表达,蛋白免疫印迹发现PEDF及受体蛋白质相对分子质量分别为50KD和55KD。PEDF主要表达在细胞浆内,PEDF受体主要表达在胞膜上。10μmol/L和100μmol/L DHT作用48h后,SZ95细胞PEDF mRNA和蛋白水平明显降低。结论在mRNA及蛋白质水平,SZ95细胞均表达PEDF及受体,雄激素可降低SZ95细胞PEDF表达。 Objective To investigate the expression of pigment epithelium derived factor(PEDF) and its receptor in a sebaceous gland cell line of SZ95. Methods RTPCR and Western blot were used to measure the mRNA and protein expressions of PEDF and its receptor,and the location was detected by immunofluorescence. Different concentrations of dihydrotestosterone(DHT) were acted on SZ95 cells as stimulating factors. Real-time quantitative PCR(q PCR) and western blot were used to measure the changes of PEDF mRNA and protein levels. Results Both the mRNA and protein expressions of PEDF and its receptor were detected in SZ95 cells. The molecular weights of PEDF and its receptor were 50 kD and 55 kD respectively. PEDF were observed mainly in the cytoplasm,and PEDF receptor on themembrane. After 48 hours of exposure to 10 μmol/L and 100μmol/L DHT,PEDF mRNA and protein levels significantly decreased. Conclusion PEDF and its receptor are expressed in SZ95 cells,and PEDF expression can be inhibited by androgen.
