摘要
为探讨蓝莓花色苷对淋巴细胞和腹腔巨噬细胞的免疫调节作用。无菌分离淋巴细胞和腹腔巨噬细胞,制备淋巴细胞和腹腔巨噬细胞悬液,以终浓度为4、8、16、32、64 mg/m L的蓝莓花色苷作用于体外培养的淋巴细胞(48 h)和腹腔巨噬细胞(24 h),采用MTT法检测蓝莓花色苷对淋巴细胞增殖率及中性红吞噬实验检测腹腔巨噬细胞吞噬功能的影响,ELISA法检测蓝莓花色苷对淋巴细胞(IL-6白细胞介素-6)、(TNF-α肿瘤坏死因子-α)和腹腔巨噬细胞(IL-4白细胞介素-4)、(IL-12白细胞介素-12)分泌的影响。结果显示,蓝莓花色苷各浓度组淋巴细胞增殖率、腹腔巨噬细胞的吞噬活性、培养上清中淋巴细胞IL-6、TNF-α及腹腔巨噬细胞IL-12的分泌高于空白组(p <0.05),各浓度组与阳性组腹腔巨噬细胞IL-4分泌量均低于空白对照组(p <0.05)。蓝莓花色苷可能通过促进淋巴细胞增殖和提高腹腔巨噬细胞吞噬及分泌功能,提高机体免疫功能。
To investigate the immunomodulatory effects of blueberry anthocyanins on lymphocytes and peritoneal macrophages.The aseptically isolate lymphocytes and peritoneal macrophages were used for preparation of the lymphocytes and peritoneal macrophages suspension.Blueberry anthocyanins at 4,8,16,32 and 64 mg/m L were used to culture lymphocytes( 48 h) and peritoneal macrophages( 24 h) in vitro. The effects of blueberry anthocyanins on the proliferation of lymphocytes and the phagocytosis of macrophages by neutral red-phagocytosis were detected by MTT method. The effects of anthocyanins on the secretion of IL-6( interleukin-6) and TNF-α( TNF-α) and peritoneal macrophages IL-4( interleukin-4),IL-12( interleukin-12) were detected by ELISA method.The results showed that the proliferation rate of lymphocytes,the phagocytic activity of peritoneal macrophages,the secretion of IL-6 and TNF-α in lymphocytes and the secretion of IL-12 in the peritoneal macrophages were significantly increased compared with that in the blank group( p〈0.05) but the secretion of IL-4 in peritoneal macrophages of each concentration group and positive group was lower than that of the blank control group( p〈0.05).The blueberry anthocyanins may enhance the immune function of human body by promoting the proliferation of lymphocytes and increasing the phagocytic and secretory functions of peritoneal macrophages.
