摘要
目的:观察不同浓度前列腺素E_2受体2(EP2)抑制剂AH6809对BV-2细胞的致凋亡作用,探索AH6809使用浓度与BV-2细胞凋亡的关系。方法:将不同浓度(10、20、30、40、80、160μmol/L)的AH6809依次加入BV-2细胞培养液中,运用倒置显微镜、免疫荧光双标、TUNEL检测、流式细胞术和Western Blot技术,对AH6809干预后的BV-2细胞的形态和凋亡情况进行观察。结果:AH6809在40μmol/L以上时,观察到BV-2细胞开始出现凋亡迹象,并且随着AH6809浓度的递增,发生凋亡的BV-2细胞呈现出逐渐增多的趋势,表现出浓度依赖性特征,差异具有统计学意义(P <0. 05)。结论:AH6809对BV-2细胞有致凋亡作用,使用时需要考虑浓度对细胞凋亡的影响。
Objective: To observe the apoptosis-inducing effect of prostaglandin E2 receptor 2( EP2) antagonist AH6809 at different concentrations on BV-2 cells and explore the relationship between the concentration of AH6809 and the apoptosis of BV-2 cells.Methods: Different concentrations of AH6809( 10,20,30,40,80 and 160 μmol/L) were respectively added to the BV-2 cells culture medium.After the intervention of AH6809,inverted microscope,doublelabeling immunofluorescence,TUNEL assay,flow cytometry and Western Blot were used to observe the change of morphology and the apoptosis of BV-2 cells.Results: The results showed that when the concentration of AH6809 was above40 μmol/L,BV-2 cells began to show the signs of apoptosis.With enhancing concentration of AH6809,the apoptosis of BV-2 cells showed a gradually increasing trend and concentration-dependent characteristics.The difference was statistically significant( P〈0.05).Conclusion: AH6809 shows the apoptosis-inducing effect on BV-2 cells,and it is necessary to consider the effect of the concentration on the apoptosis of the cells.
