福建省一株猪瘟病毒流行毒株全基因组序列测定与分析 被引量:2

Genome Sequence Analysis of a Classical Swine Fever Virus Isolate from Fujian Province

摘要 根据GenBank公布的猪瘟病毒(CSFV)标准参考毒株全基因组序列,设计11对引物,应用RTPCR方法分别对11个片段进行福建流行毒株CN-FJLY的全基因组扩增,与参考毒株进行序列分析比较。结果表明,福建流行毒株CN-FJLY基因组全长为12 296bp,与参考毒株SXCDK、HEBZ、GXWZ02、HNLY-2011和Zj0801的全基因组核苷酸同源性为93.0%~97.0%,而与1.1亚群的中国强毒Shimen、疫苗株HCLV全基因组核苷酸同源性为84.4%~85.3%。基于全基因组及E2基因构建的遗传进化树分析结果表明福建流行毒株CN-FJLY属于2.1b亚群,与Shimen株、HCLV疫苗株亲缘关系较远。与HCLV相比较CN-FJLY基因组变异较大的是5′UTR、Ems、E2、p7、NS2、NS5A、NS5B和3′UTR。综上说明福建省猪瘟病毒流行毒株正在向远离疫苗株的方向变异。 One CSFV strain named CN-FJLY isolated from Fujian province was sequenced with 11 pairs of primers based on CSFV standard reference strain genome sequences in GenBar-k. Sequencing of the com- plete genome of CN-FJLY showed that the genome length was 12296 bp,and shared 93.0%-97.0- with SXCDK,HEBZ,GXWZ02,HNLY-2011 and Zj0801,but only 84.4%-85.3% homology to the Shimen and HCLV strain. Phylogenetic analysis based on complete genom'e and E2 gene showed that CN-FJLY was classified into subgenotype 2.1b.In addition,we found that 5 'UTR,Ems,E2,P7,NS2,NSSA,NSSB and 3' UTR were the most variable genes in CN-FJLY compared to HCLV. In summary, the epidemic strain of CSFV in Fujian province was highly variable in comparing to vaccine strain.
