防腐生肌膏对基质金属蛋白酶变化的影响 被引量:1

摘要 通过防腐生肌膏对创面肉芽组织中基质金属蛋白酶(MMPs)及其抑制剂(TIMPs)表达调控,阐明其促进肉芽组织增生的机制。试验以SD大鼠制备皮肤创伤模型,给药后分不同时间点取创面肉芽组织,采用ELISA检测创缘组织中MMP-1、MMP-2、MMP-9及TIMP-1和TIMP-2的含量。结果显示,创伤后第3天,防腐生肌膏组创面MMP-1的表达量达到最高,与对照组比较差异显著(P<0.05);创伤后防腐生肌膏组创面MMP-2表达量逐渐增加,第10天达到高峰,与对照组差异显著(P<0.05);防腐生肌膏能极显著促进创面表达MMP-9(P<0.01),创伤后第3天达到表达高峰。防腐生肌膏组与对照组比较,TIMP-1、TIMP-2的表达量逐渐减少,并且极显著低于对照组(P<0.01)。综上所述,防腐生肌膏能通过促进创面MMP-1、MMP-2、MMP-9的表达,抑制创面TIMP-1、TIMP-2的表达来调节胶原代谢促进创伤愈合。 The study was to regulate the expressions of matrix metalloproteinases(MMPs) and their tissue inhibitor of matrix metalloproteinases (TIMPs) in the granulation tissue of the wound through the antisep- tic spermatin and to elucidate its mechanism of promoting granulation tissue hyperplasia.The skin wound model was prepared in SD rats, and granulation tissues were taken at different time points after administra- tion.The contents of MMP-1 ,MMP-2 ,MMP-9 ,TIMP-1 and TIMP-2 in the wound tissues were detected by ELISA.The expression of MMP-1 in the wound of Fangfu Shengji ointment group was the highest at the third day after trauma,and the difference was significant (P-0.05).The expression quantity of MMP-2 was gradually increased,and reached the peak at the 10th day,which was significantly different from the control group.Fangfu Shengji ointment could significantly promote the expression of MMP-9 (P%0.01) in wound,and reached the peak of expression third days after trauma. Compared with the control group,the expression of TIMP-1 and TIMP-2 gradually decreased, and significantly lower than that in the control group (P%0.01).In summary,Fangfu Shengji ointment can promote the expressions of MMP-1,MMP-2 and MMP-9,inhibit the expressions of TIMP-1 and TIMP-2 in the wound to regulate collagen metabolism to promote wound healing.
