摘要
目的探讨MiR-223在宫颈癌细胞Hela中是否通过上皮间质转化(EMT)来抑制宫颈癌细胞的侵袭转移。方法在宫颈癌细胞Hela中过表达MiR-223,利用平板克隆实验观察转染10 d后宫颈癌细胞的克隆情况;进一步利用细胞划痕愈合实验和Transwell实验观察宫颈癌细胞的迁移侵袭能力;利用Western blot实验检测上皮间质标志物蛋白波形蛋白(Vimentin)、N-钙粘蛋白(N-cadherin)、E-钙粘蛋白(E-cadherin)和转录因子Twist的表达情况。结果在Hela细胞中过表达MiR-223后,平板克隆实验显示实验组细胞克隆数明显少于对照组,细胞生长受到抑制;划痕实验显示过表达MiR-223 24 h和48 h后,实验组迁移距离远远小于对照组,细胞迁移能力受到抑制;Transwell实验结果表明实验组细胞穿膜数量远小于对照组,细胞侵袭能力同样受到抑制。Western blot结果提示过表达MiR-223后实验组中波形蛋白、N-钙粘蛋白、Twist表达较对照组降低,E-钙粘蛋白的表达则较对照组升高。结论过表达MiR-223通过抑制上皮细胞向间质细胞转化来抑制Hela细胞侵袭迁移能力。
Objective To investigate whether MiR-223 inhibits the invasion and metastasis of cervical cancer cells byepithelial interstitial transformation(EMT) in HeLa cells. Methods After the overexpressing MiR-223 in HeLa cells, the cellproliferation was observed after 10 days by cell colony formation assays. The migration and invasion of cervical cancer cellswere tested by wound-healing assay and Transwell migration assay; and the expression of EMT-related proteins such asVimentin, N-cadherin, E-cadherin and transcription factor Twist were detected by Western blot. Results After theoverexpressing MiR-223 in HeLa cells, the cell proliferation was inhibited, the results of wound healing assay showed that themigration distance of the test group was significantly shorter than that of the control group after 24h and 48h; Transwell resultsshowed that the amount of cells through membrane was significantly less than that of the control group; Western blot resultsshowed that the expression of E-cadherin increased in the test group, the expression of N-cadherin, Vimentin and Twistdecreased. Conclusions The overexpressing MiR-223 can inhibit the migration and invasion in HeLa cells throughinhibiting transform the epithelial cells into the mesenchymal cells.
