摘要
目的:考察益母草不同活性组分对人脐静脉血管内皮细胞(HUVECS)增殖以及斑马鱼血管生成的作用。方法:运用MTT法测定益母草乙酸乙酯部位和正丁醇部位对HUVECS增殖的影响。采用斑马鱼观察益母草乙酸乙酯部位和正丁醇部位对斑马鱼体节间血管生成的影响。结果:益母草乙酸乙酯部位在3.125~50μg/mL浓度范围对HUVECS细胞具有一定的损伤作用,在6.25~50μg/mL范围可明显抑制斑马鱼体节间血管生成,体节间血管指数显著下降(P<0.01);益母草正丁醇部位在12.5~50μg/mL范围对sunitinib诱导的HUVECS细胞损伤具有一定的保护作用(P<0.01)。在6.25~50μg/mL范围对sunitinib诱导的斑马鱼体节间血管损伤具有明显保护作用,与模型组比较,血管指数上升(P<0.05,P<0.01)。结论:益母草对血管生成具有双向调节作用,与其不同的药效物质密切相关。
Objective: To investigate the effects of different extraction parts of Leonurusjaponicus on human umbilical vein endothelial ceils (HUVECs) proliferation and zebrafish angiogenesis. Methods: The effects on HUVECs proliferation of ethyl acetate extract and n-butyl alcohol extract of L. japonicus were investigated by MTT method. The effects on intersegment vessels (ISVs) formation in zebrafish of ethyl acetate extract and n-butyl alcohol extract of L. japonicus were observed under fluorescent microscope. Results: At concentrations between 3.125 and 501μg/mL, ethyl acetate extract could injure HUVECs significantly. Meanwhile, 6.25-501μg/mL of ethyl acetate extract could inhibit ISVs angiogenesis, and ISV index decreased significantly (P〈0.01 ). In contrast, 12.5-501μg/mL of n-butyl alcohol extract could repair the damage of HUVECs induced by sunitinib (P〈0.01). 6.25-501μg/mL of n-butyl alcohol extract could repair the damage of intersegment vessels induced by sunitinib, when compared to the model group (snnitinib), ISV index increased significantly (P〈0.05, P〈0.01). Conclusion: L. japonicus has biphasic effect on angiogenesis, which is closely associated with the different effective materials.
