五味苦参肠溶胶囊对溃疡性结肠炎小鼠炎性因子的影响 被引量:13

Effects of Five-Flavor Sophora flavescens Enteric-coated Capsules on inflammatory factors in mice with ulcerative colitis

摘要 目的:观察新药五味苦参肠溶胶囊(FSEC)对葡聚糖硫酸钠(DSS)诱导的溃疡性结肠炎(UC)小鼠维甲酸相关孤儿受体γ-t(RORγt)、白介素(IL)-17、IL-6浓度及髓过氧化物酶(MPO)活性的影响。方法:BALB/C小鼠50只,随机分成5组:空白组(Nctrl),模型组(Model),FSEC低、中、高剂量组(1.92、3.84、7.68g·kg^(-1)·d^(-1)),每组10只。除空白组外,其余各组自由饮用3%DSS 7d,然后空白组与模型组0.9%氯化钠溶液灌肠,FSEC各剂量组分别按不同剂量灌肠7d。各组取结肠组织观察炎症损伤评分(DAI),HE染色光镜下观察结肠组织病理变化及ELISA法检测结肠组织中RORγt、IL-17、IL-6蛋白含量及MPO活性。结果:HE染色观察,与空白组比较,模型组结肠组织炎症损伤严重,而FSEC各组炎症损伤均有不同程度的改善。与模型组比较,FSEC各剂量组小鼠结肠组织中RORγt、IL-17、IL-6炎性因子及MPO活性的表达均有所降低(P﹤0.05,P﹤0.01),其中FSEC中剂量组效果最显著。结论:FSEC可降低DSS诱导的UC小鼠结肠组织中RORγt、IL-17、IL-6蛋白水平及MPO活性,减轻结肠组织的病理损伤,缓解UC小鼠的症状。 Objective: To observe the effects of Five-Flavor Sophora flavescens Enteric-coated Capsules (FSEC) on the protein level of retinoid-related orphan receptor gamma-t (ROR γ t), interleukin-17 (IL-17) and interleukin-6 (IL-6) and the activity of myeloperoxidase (MPO) in ulcerative colitis (UC) mice induced with dextran sulfate sodium (DSS). Methods: Fifty BALB/C mice were randomly divided into five groups: normal control group (Nctrl group), model control group (Model group), low, middle, high dose (1.92, 3.84, 7.68 g.kg^-l·d^-1) of FSEC group with ten mice per group. Except Nctrl group, all groups received 3% DSS drinking water for seven days, then Nctrl group and model group had 0.9% saline enema for seven days, while FSEC groups had FSEC suspension enema of different concentration respectively as mentioned before for seven days. The scores of disease activity index (DAI) and the activity of myeloperoxidase (MPO) were measured. The pathology of colon tissues was stained by HE and observed under the light of microscope. The protein level of RORγt, IL-17 and IL-6 were detected by ELISA. Results: HE staining: compared with Nctrl group the model group's colon tissue structure was disordered, while the inflammation was reduced by different degree in FSEC groups. ELISA results: Compared with the model group, FSEC groups downregulated of the protein level of RORγt, IL-17, IL-6 and MPO activity (P〈0.01, P〈0.05), the middle dose of FSEC group had significant effect. Conclusion: FSEC can reduce the protein level of RORγt, IL-17, IL-6 and MPO activity in mice with DSS-induced UC, alleviate the pathological injury of gut, and relieve symptoms.
