摘要
目的:通过观察蒙药齐顺保利尔对造血、凝血功能的影响,揭示其主要药效学作用.方法:本研究由蒙药齐顺保利尔对正常小鼠骨髓造血功能的影响、对60 Co-g射线照射所致小鼠骨髓造血功能损伤的影响、对环磷酰胺所致小鼠骨髓造血功能损伤的影响等三项实验构成.各实验均将小鼠随机分入正常对照组、模型组、血康胶囊组、齐顺保利尔高、中、低剂量组;以60Co-g射线照射(Co-g Ray Radiation)和腹腔注射环磷酰胺等两种方法制备骨髓抑制模型,并给予0.5%CMCNa、血康胶囊或齐顺保利尔高、中、低剂量溶液,取静脉血和骨髓,检测外周血细胞和骨髓有核细胞变化,观察小鼠断尾出血时间的影响,并采用玻片法检测凝血时间.结果:实验发现齐顺保利尔对正常小鼠骨髓造血功能无明显影响;齐顺保利尔高、中、低剂量对60Co-g射线照射所致小鼠WBC、PLT、骨髓有核细胞的降低均有明显的升高作用,且呈现量效关系;齐顺保利尔可显著降低细胞毒剂—环磷酰胺的细胞毒作用,促进骨髓有核细胞的分化和增殖,从而使外周血WBC、PLT数增加;齐顺保利尔可明显缩短小鼠断尾出血时间、明显缩短小鼠凝血时间.结论:齐顺保利尔具有促进骨髓造血功能、止血功能.
Objective: Through observing the effects of Mongolian medicine Qishunbaolier on hematopoiesis and coagulation function, to reveal its main pharmacodynamic effects. Methods: The effects of Qishunbaolier on hematopoieticfunction of normal mouse, radiation-induced damaged mice, and cyclophosphamide-induced bone marrow damaged mice were determined. Mice were randomly divided into a normal control group, a model group, a Xuekang capsule group, and Qishunbaolier groups with high, middle and low dosages. 60Co-ray irradiation(Co-Ray Radiation)and intraperitoneal injection of cyclophosphamide were used to prepare the mouse myelosuppressive model.CMCNa(0.5%), Xuekang capsule or high, medium and low doses of Qishunbaolie solutions were administered astreatments. The mouse venous blood and bone marrow were used to detect the changes of peripheral blood cells and bone marrow nucleated cells, The bleeding time of mouse severed tail was observed, and the blood clotting time was measured by glass slide method. Results: Qishunbaolier had no significant effect on the bone marrow hematopoietic function of normal mice. Qishunbaolier with high, middle and low dosages had inhibiting effects on the decrease of WBC, PLT and bone marrow nucleated cells caused by 60Co-ray irradiation. There was a dose-effect relationship.Qishunbaolier could significantly reduce the cytotoxicity of cytotoxic agent-cyclophosphamide and promote the proliferation and differentiation of bone marrow nucleated cells, thereby increasing the number of WBC and PLT in peripheral blood. Qishunbaolier could significantly shorten the bleeding time of severed tail and the blood clotting time.Conclusion: The main pharmacological effects of traditional Mongolian medicine Qishunbaolier are promoting hematopoiesis and coagulation.
