摘要
应用低浓度美兰染色液(2.0%)对原生动物进行梯度微麻醉,麻醉后的虫体活动性减弱但伸缩泡活动状态良好,能够较清晰的观察伸缩泡及收集管的交替活动,可计数活动频率,且这种状态的维持时间较长(约19min)。为将细胞压制为既较薄又不破碎,实验应用低浓度(1%)甲基纤维素作为支撑物,结果表明,该方法操作简单,效果较好,适用于高校或中学动物学实验的课堂操作。
Protozoa was carried out with low concentration of methylene blue staining solution (2.0%) to be gradient micro-anesthetized.The activity of the insects after anesthesia was weakened but the activity of the contractile vacu- ole was good.It was possible to observe the alternating aetivities of the contractile vacuole and the collecting tube more clearly.The activity frequency can be counted.This state is maintained for a longer period of time (about 19min- utes).To suppress the cells to be both thin and not broken, the experiment used low eoncentrations(1%) of methyl cellulose as a support.This method has a simple operation and is suitable for classroom operation in laboratory experi- ments in middle schools or colleges.
