老龄大鼠血清非对称性二甲基精氨酸浓度变化及其与心肌细胞衰老的关系

摘要 目的探讨老龄大鼠血清中非对称性二甲基精氨酸(ADMA)浓度的变化情况及其与心肌细胞衰老的关系。方法选择23月龄Wistar大鼠作为老年组,4月龄Wistar大鼠作为青年组,测定血清ADMA和血脂水平、心肌组织中二甲基精氨酸二甲胺水解酶(DDAH)活性、一氧化氮合酶(NOS)活性和一氧化氮(NO)含量,RT-PCR法测定心肌组织中P21 mRNA、P53 mRNA、Sirt1 mRNA水平。结果老年组血清ADMA水平明显高于青年组(P<0. 05),空腹血糖和胰岛素水平低于青年组(P<0. 05),血清总胆固醇、低密度脂蛋白水平高于青年组(P<0. 05),血清高密度脂蛋白水平低于青年组(P<0. 05),两组血清三酰甘油水平比较差异无统计学意义(P>0. 05)。老年组心肌组织中DDAH活性、NOS活性和NO含量低于青年组(P<0. 05); P21 mRNA和P53 mRNA水平高于青年组(P<0. 05),Sirt1 mRNA水平低于青年组(P<0. 05)。老年组血清ADMA水平与心肌组织中P21 mRNA、P53 mRNA水平呈正相关(P<0. 05),与心肌组织中Sirt1 mRNA水平呈负相关(P<0. 05)。结论老龄大鼠血清中ADMA水平升高,ADMA水平和心肌细胞衰老有关,其机制可能与ADMA减少NO生成、降低P21和P53水平、升高Sirt1水平有关。
