摘要
目的探讨不同品系小鼠颈部心脏移植后Th1/Th2细胞因子的变化及意义。方法建立小鼠颈部心脏异位移植模型,实验分3组,即同系移植组、同种移植组和同种环孢素A(CsA)组。比较3组供心存活时间、排斥反应程度,流式细胞仪检测供心血清中Th1/Th2细胞因子变化。结果同系移植组移植心脏存活时间最长,移植后排斥反应最轻。术后第7天,同种移植组Th1/Th2表达水平明显高于同系移植组和同种CsA组(P<0.05)。结论 Th1/Th2细胞因子变化与心脏移植后排斥反应的发生发展密切相关,可作为心脏移植后排斥反应的监测指标。
Objective To investigate the changes of Th1/Th2 cytokines after cervical heart transplantation in different strains of mice and their significance. Methods The mice models of heterotopic cervical heart transplantation were established. All the mice were divided into three groups, the syngeneic transplantation group, allograft trans-plantation group, and the cyclosporine A(CsA)-treated allograft transplantation group. The survival time and rejection degree of the donor hearts were compared among the three groups. Flow cytometry was used to determine the changes of Th1/Th2 cytokines in the serum of donor hearts. Results The transplanted hearts in the syngeneic group showed the longest survival time and the least rejection degree after transplantation. At 7 d after the operation, the expression level of Th1/Th2 cytokines in the allograft transplantation group was significantly higher than that in the syngeneic group and the CsA-treated allograft transplantation group(P〈0.05). Conclusion The changes of Th1/Th2 cytokines are closely related to the occurrence and development of rejection after heart transplantation. It can be used as a mon-itoring index for rejection after heart transplantation.
