补骨脂中补骨脂素和异补骨脂素提取工艺研究 被引量:5

摘要 目的:采用正交试验设计、星点设计-效应面法优化补骨脂中补骨脂素和异补骨脂素的提取工艺。方法:分别采用正交试验设计与星点设计-效应面法,以乙醇浓度、乙醇用量、提取时间、提取次数为自变量,补骨脂素和异补骨脂素的总提取率作为指标,优化提取工艺。结果:正交试验设计确定的最佳提取工艺为10倍量体积分数60%乙醇回流提取3次,每次1 h;星点设计-效应面法确定的最佳提取工艺为11.15倍量体积分数55.7%的乙醇回流提取3次,每次1.15 h。结论:两种方法优化的提取工艺相似。正交试验设计实验次数少,但精确度不高;星点设计-效应面法实验次数多,但其预测性良好,精确度更高。为得到更高的精确度,并尽量减少实验次数,可将正交试验与星点设计-效应面法联用,优化药物提取工艺。 Objective:To optimize the process of extacting effective constituents of coumarin principal constituents psoralen and isopsoralen from Psoralea corylifolia L. by orthogonal experimental design and central composite design-reponse surface method. Methods:Orthogonal experimental design and star point design-effect surface method were used respectively and the concentration of ethanol, consumption of alcohol, extraction time, extraction times were used as independent variables. The total extraction rate of psoralen and isopsoralen from Psoralea corylifolia were used as indicators to optimized the extraction process. Results : The optimum extraction process determined by orthogonal experimental design was 60 % ethanol 60 minutes for reflux 10 fold of solvent and 3 times for extraction. The best extraction process determined by the star point design effect surface optimization method was 55.7 % ethanol 69 minutes for reflux 11.15 fold of solvent and 3 times for extraction. Conclusion:The extraction process optimized by the two methods is similar. The number of experiments in orthogonal design is small, but the accuracy is not high. The number of stars in the design of the star point design-effect surface method is large, but its predictability is good and the accuracy is higher. In or- der to obtain higher accuracy and minimize the number of experiments, the orthogonal test can be combined with the star point de- sign-effect surface optimization method to optimize the drug extraction process.
