摘要
目的:比较桑叶固态发酵前后槲皮素和山奈酚的含量变化。方法:色谱柱为Shim-pack VP-ODS C18(4.6 mm×250.0mm,5μm),流动相为乙腈-(体积分数)0.1%磷酸(20∶80);流速为1 mL·min^(-1);柱温设定为30℃;进样量为10μL;检测波长为368 nm;采用药用真菌固态发酵技术对桑叶进行发酵,然后利用高效液相色谱法测定发酵前后桑叶样品中槲皮素和山奈酚的含量。结果:槲皮素和山奈酚分别在0.066 8~1.002 0μg和0.038 2~0.573 0μg范围内线性关系良好,平均加样回收率分别为98.38%(RSD=1.16%)、98.42%(RSD=1.72%)。发酵后,桑叶中槲皮素和山奈酚的含量之和较发酵前提高了31.87%。结论:采用药用真菌固态发酵技术发酵桑叶,能提高槲皮素和山奈酚的含量。
Objective : To compare the changes of quercetin and kaempferol before and after fermentation of Chinese mulberry leaves (Folium moil) in solid state. Method:The chromatogram column was Shim-pack VP-ODS C18 (4.6 mm × 250 mm,5 μm), and the mobile phase was acetonitrile-( volume fraction) 0.1% phosphoric acid (20 : 80). The flow rate was 1 mL · min^-1 and the column temperature was set to 30℃. The injection volume was 10 μL and the detection wavelength was 368 nm. The mulberry leaves( Folium moil) were fermented by medicinal fungal solid-state fermentation technology,and then the high-performance liquid chromatography was used to determine the sputum samples in the traditional Chinese medicine mulberry leaves before and after fer- mentation. The content of quercetin and kaempferol in Chinese mulberry leaves samples before and after fermentation was deter- mined by high performance liquid chromatography. Results:The linear relationship between quercetin and kaempferol was in the range of O. 066 8 to 1. 002 0 μg and 0.038 2 to 0.573 0 txg,respectively. The average recovery was 98.38% ( RSD = 1.16% ) and 98.42% (RSD = 1.72% ). After fermentation, the contents of quercetin and kaempferol in mulberry leaves increased by 31. 87% compared with that before fermentation. Conclusion:Fermentation of mulberry leaves(Folium moil) by solid fermentation of medicinal fungi can increase the content of quercetin and kaempferol.
