摘要
目的检测牙髓组织中肌腱及成牙本质相关分子的表达。方法采用组织学染色对牙髓组织进行形态学观察,同时运用逆转录PCR (RT-PCR)、免疫荧光等方法检测牙髓组织中肌腱及成牙本质相关分子的表达。结果组织学观察到牙髓组织分为成牙本质细胞层、乏细胞层、多细胞层与髓核四层,并含有胶原纤维和肌纤维。RT-PCR发现牙髓组织明显表达肌腱及成牙本质相关基因,如Six1、TNC、Col-Ⅰ、Col-Ⅲ、DSPP、Runx2、BSP、OPN、OCN等基因,低表达SCX、TNMD、Six2、DMP1基因。免疫荧光发现在牙髓组织的成牙本质细胞层明显表达DSPP和SCX蛋白。结论牙髓组织中的细胞具有向肌腱、成牙本质相关组织分化的潜能。
Objective To detect the expression of tendon and odontogenesis associated markers in dental pulp tissue. Methods Histological staining was used to observe the morphology of dental pulp tissue. NT-PCN and immunofluorescence were applied to detect the expression of tendon and odontogenesis related molecules in dental pulp tissue. Results Histologically, dental pulp tissue was divided into four layers: odontoblastic layer, cell free zone, cell rich zone and pulp core, and some coIIa gen fibers and muscle fibers were found in it. The expression of tendon and odontogenesis associated genes, such as Sineoculis homeobox homolog (Six) 1 , Tenascin C (TNC) , Collagen-Ⅰ (Col-Ⅰ) , Collagen-Ⅲ(Col-Ⅲ) , dentin sialophosphoprotein (DSPP) , Runt related transcription factor 2 (Runx2) , Bone sialoprotein (BSP) , osteopontin (OPN) , osteocalcin (OCN) were obviously detected in the pulp tissue by RT PCR, while the genes of scleraxis (SCX) , tenomodulin (TNMD), Six2 and dentin matrix protein (DMP) 1, were lowly expressed, Immunofluorescent expressed in the odontoblastic layer of dental pulp tissue. Conclusion ate into tendon and odontogenesis related tissues. results revealed that DSPP and SCX proteins were clearly Cells in dental pulp tissue have the potential to differentiate into tendon and odontogcncsis related tissues.
