摘要
以大花六道木幼嫩叶片为外植体,研究不同植物生长调节剂对不定芽诱导的影响,并采用正交试验设计的方法,研究基本培养基、生长素和蔗糖浓度对组培苗生根的影响。结果表明:诱导大花六道木不定芽发生的最适培养基为改良MS,同时添加0. 4 mg/L 6-BA、30 g/L蔗糖和2. 8 g/L结冷胶,在黑暗条件培养1个月后,不定芽直接从叶片伤口处发生,诱导率在35%左右。生根培养以WPM或MS为基本培养基,同时添加0. 5 mg/L IBA、1. 0 mg/L NAA、20 g/L蔗糖和7 g/L琼脂粉,生根率在90%左右。本研究建立了一种大花六道木不定芽离体再生的方法,可用于大花六道木的快速规模化繁殖及遗传改良。
We used the lamina explants of Abelia grandiflora to investigate the effects of various plant growth regulators on shoots regeneration. Also, the effects of basal mediums, auxin and sucrose on rooting of buds were investigated by orthogonal design statistic analysis. The resuh showed that, after one month cultured in dark, the adventitious shoots were directly regenerated from lamina explants on modified MS medium, supplemented with 0.4 mg/L 6-BA, 30 g/L sucrose and 2.8 g/L geMte, and the percentage of explants responding to the induction amounted to 35%. Up to 90% of shoots rooted on WPM or MS medium supplemented with 0.5 mg/L IBA, 1.0 mg/L NAA,20 g/L sucrose and 7 g/L agar. In this study, a method of adventifous shoots inducfon from in vitro cultured leaves of Abelia grandiflora was established, which could be applied to mass propagation and improvement for Abelia grandiflora.
