振动暴露时间对悬浮红细胞质量影响初探 被引量:5

摘要 目的模拟极端路况下运输悬浮红细胞,研究不同时长振动对悬浮红细胞质量的影响。方法使用电磁振动实验系统模拟极端陆地运输条件下的振动,选择组合轮式车振动环境,水平、纵向、垂直3方向振动,将悬浮红细胞累计振动6 h。分别在振动前、累计振动3 h、4. 5 h、6 h后留取血样,检测游离血红蛋白(FHb)、乳酸脱氢酶(LDH)、钾离子(K~+)、溶血率,分析比较振动前后及不同振动暴露时间对上述指标的影响。结果悬浮红细胞FHb含量在振动3 h、4. 5 h、6 h分别为1. 30±0. 42 g/L、1. 66±0. 46 g/L和2. 14±0. 54 g/L与振动前(0. 28±0. 06 g/L)相比有显著差异(P〈0. 05);振动后溶血率较振动前明显增高,振动3 h、4. 5 h、6 h分别为0. 25%±0. 07%、0. 36%±0. 07%和0. 45%±0. 11%,与振动前(0. 04%±0. 02%)相比较有显著差异,P〈0. 05;振动后LDH较振动前明显增高,振动3h、4. 5 h、6 h分别为(507. 20±134. 70) U/L、(679. 40±151. 10) U/L和(838. 50±169. 90) U/L与振动前[(69. 4±27. 9) U/L)比较有显著差别,P〈0. 05;振动3 h、4. 5 h、6 h K~+浓度分别为(9. 6±0. 72) mmol/L、(10. 2±0. 84) mmol/L和(10. 6±0. 84) mmol/L均高于振动前K~+浓度[(6. 9±0. 56) mmol/L],P〈0. 05,振动4. 5 h及振动6 h后的K~+较振动3 h后增高(P〈0. 05),但振动6 h后与振动4. 5 h后的K~+比较差异无统计学意义(P〉0. 05)。结论剧烈的陆地运输振动可导致悬浮红细胞FHb、溶血率、LDH及K~+含量的改变,导致红细胞振动损伤,且FHb、溶血率及LDH随着振动暴露时间的延长而逐渐增高,影响悬浮红细胞质量。 Objective To investigate the effect of vibration exposure duration on the quality of red blood cells( RBCs)during transportation. Methods An electromagnetic vibration test platform was used to stimulate the transportation environment under case of military wheeled vehicles( three direction vibration of horizontal,longitudinal,vertical). Suspended erythrocytes were vibrated for a total of 6 hours. Samples were collected before/after the vibration and 3 h,4. 5 h,6 h into the vibration. Free Hemoglobin( FHb),K~+,LDH and hemolysis ratio were measured. Results The FHb,K~+,LDH and hemolysis ratio of the RBCs were significantly increased after vibration compared to the initial status( P〈0. 05). The FHb increased as the exposure time of vibration prolonged( 0. 28±0. 06 g/L vs 1. 30±0. 42 g/L vs 1. 66±0. 46 g/L vs 2. 14±0. 54 g/L,P〈0. 05). The same trend was observed in LDH level( 69. 40± 27. 90 U/Lvs 507. 20± 134. 70 U/L vs 679. 40± 151. 10 U/L vs 838. 50±169. 90 U/L,P〈0. 05). The concentrations of K~+at 4. 5 h and 6 h into the vibration were higher than that at 3 h( P〈0. 05). However,there was no statistically significant difference in the concentrations of K~+between 4. 5 h and 6 h( P〉0. 05). Conclusion The FHb,K~+,LDH and hemolysis ratio of the suspended erythrocytes were significantly increased after a 6 h vibration period. Based on our data,the increase of FHb,hemolysis ratio and LDH is related with the exposure duration to the vibration.
