摘要
前期研究表明,茄子Enhanced disease susceptibility 1(SmEDS1)正调控茄子青枯病抗性,但是其抗病机理有待进一步研究。为筛选SmEDS1的互作蛋白,本研究构建pGBK7-SmEDS1诱饵载体,并利用酵母双杂交技术对相应的均一化cDNA酵母文库进行筛选。从2次独立的实验中筛选到9个克隆,其非冗余地编码6种蛋白,即TCP转录因子、DNAJ分子伴侣、SnRK1蛋白α亚基、氰酸酯酶、CIP4蛋白和1个未知蛋白。基于这些蛋白的功能,推测SmEDS1可能通过与本研究筛选到TCP转录因子互作调控水杨酸的合成,进而调控茄子的青枯病抗性。
In our previous study, EDS1 of eggplant(SmEDS1) was found positively regulating the bacterial wilt resistance, but the resistance mechanism was not known. To screen the candidate interactors of SmEDS1, the bait protein vector of pGBK7-SmEDS1 was construct to screen the cDNA library by Y2 H. The result showed that total nine clones were obtained by two independent experiments. The nine clones encode six proteins including TCP transcriptional factor, DNAJ molecular chaperone, cyanate hydratase, CONSTANS interacting protein 4 and an unknown protein. Based on the known function of the candidate protein, SmEDS1 interacted with the TCP, which regulating the synthesis of salicylic acid and finally regulating the bacterial resistance.
