摘要
丙酮酸激酶有4种同工酶,其中活性最强的M2型丙酮酸激酶(PKM2)是糖酵解途径中的限速酶,它在催化磷酸烯醇式丙酮酸生成丙酮酸中起关键作用。PKM2可根据肿瘤细胞代谢的需要在低活性二聚体和高活性四聚体之间进行动态转换,从而在肿瘤代谢和增殖中起重要作用。目前,关于PKM2的调控机制尚不清楚。但通过探讨PKM2在蛋白质相互作用、微RNA、信号分子等方面对肿瘤代谢的影响,阐述PKM2和肿瘤信号通路之间的联系,挖掘PKM2作为临床肿瘤诊断和治疗靶点的潜力,可为临床恶性肿瘤的早期诊断和治疗提供新思路。
Pyruvate kinase has four isoenzymes, and the most active pyruvate kinase M2 ( PKM2 ) is the rate-limiting enzyme in the glycolytic pathway , which plays a key role in catalyzing the production of pyruvate by phosphoenolpyruvate. PKM2 can dynamically switch between low-activity dimers and high-activity tetramers according to the needs of tumor cell metabolism, thus playing an important role in tumor metabolisnl and proliferation. At present, the regulation nlechanism of PKM2 is still unclear. However, by investigating the effects of PKM2 on tumor metabolism in terms of protein interactions, microRNA, signaling molecules, the relationship between PKM2 and tumor signaling pathways is explored, and the potential of PKM2 as a clinical tumor diagnosis and therapeutic target can be explored,which can provide new ideas for the early diagnosis and treatment of malignant tumor.
